1:实验设计与方法选择:
本研究采用系统性免疫抑制的鼠模型(C3H/HeJrd1/rd1小鼠)探究人骨髓CD34+细胞玻璃体内注射的效果。方法包括活体成像(SLO/OCT)、ERG检测、免疫组化和微阵列分析。
2:样本选择与数据来源:
使用32只小鼠,人骨髓穿刺液来自健康供体。小鼠经免疫抑制后分别注射CD34+细胞或PBS。

设备包括Alzet微量渗透泵、磁性细胞分选仪(Miltenyi)、SLO/OCT成像系统、ERG系统(LKC Big Shot)、显微镜(Nikon)及微阵列工具(Affymetrix)。材料包含免疫抑制剂(他克莫司、雷帕霉素)、EGFP标记试剂、抗体及RNA提取试剂盒。
