5-ALA application can inactivate both types of bacteria, but at different rates. Further studies are needed to evaluate its effect on other types of microbes that induce dental caries because different types of bacteria are involved in development of carious lesions. In addition, in vivo studies should be carried out to determine its effect in the oral cavity.

The study was conducted in vitro, which may not fully replicate the conditions in the oral cavity. Further in vivo studies are needed to determine the effect of 5-ALA in the oral cavity.
1:Experimental Design and Method Selection
The study was designed to evaluate the antibacterial effects of 5-ALA in combination with LED light (635nm) and compared with other disinfection methods (2% CHX, 1% NaOCl) on S. mutans and S. sobrinus. The reduction in bacterial count was evaluated by the colony counting method.
2:Sample Selection and Data Sources
Sixty extracted human third molars were used. The teeth were collected from patients who had given their informed consent and stored in distilled water until use. The teeth were randomly allocated to six testing groups.
3:List of Experimental Equipment and Materials
Equipment and materials included LED light (635nm), 5-ALA, 2% CHX, 1% NaOCl, BHI agar, anaerobic jar, PCR machine, UV transilluminator, and digital camera.
4:Experimental Procedures and Operational Workflow
The teeth were prepared into dentin slabs, inoculated with S. mutans and S. sobrinus, and treated with different disinfection methods. The bacterial reduction was evaluated by the colony counting method after treatment.
5:Data Analysis Methods
The data were analyzed by counting the mean of the bacterial colonies for both positive (untreated group) and the amount of remaining viable bacteria after each disinfection method. The log reduction results were calculated by subtraction of the initial values from the final values of CFU mg–1 of dentin after being transformed by log10.
