研究目的
Investigating the therapeutic effects of a specific herbal medicine on a particular disease.
研究成果
64Cu-NOTA-Nb6 is a promising probe in NSCLC diagnosis, lesion detection and guidance of targeted therapy with PET molecular imaging. It is possible to stratify cancers and predict the prognosis of NSCLC in the clinic with 64Cu-NOTA-Nb6.
研究不足
The resolution ratio of Micro-PET images is low after 38 hours due to the decay of 64Cu, making it not appropriate to observe.
1:Experimental Design and Method Selection:
The study involved the conjugation of Nb6 with the bifunctional chelator NCS-Bz-NOTA and further labeling with the radio-nuclide 64Cu. The product was purified and its stability, binding affinity, and tumor detection ability were tested.
2:Sample Selection and Data Sources:
Human lung adenocarcinoma A549 cell lines and A549 xenografted tumor model mice were used.
3:List of Experimental Equipment and Materials:
Micro-PET equipment, PD-10 column, NanoDrop 2000 UV-Visible Spectrophotometer, MALDI-TOF-MS, Radio-TLC.
4:Experimental Procedures and Operational Workflow:
The labeling procedure, purification, stability tests, binding affinity measurement, and Micro-PET imaging were detailed.
5:Data Analysis Methods:
Data were analyzed through Graph Pad Prism Software to determine the Kd and Bmax values.
