To study the immunocorrective effects of complex treatment including the light effect of different wavelengths (red ? = 660 nm, green = 530 nm, blue ? = 440 nm) and the use of hematopoietic stem cells and platelet growth factor in patients with chronic purulent wounds of the lower extremities.

The combined use of photodynamic therapy, hematopoietic stem cells and platelet-derived growth factor is an effective treatment for septic complications, which can be considered as a new technology for correcting afferent and efferent immunoreactivity in patients with chronic purulent wounds that are not amenable to antibiotic therapy.

The study was limited to patients aged from 58 to 76 years with chronic purulent wounds of the lower extremities. The effectiveness of the treatment may vary in different age groups or with different types of wounds.
The study examined 35 patients aged from 58 to 76 years with chronic purulent wounds of the lower extremities who received targeted combined treatment: photodynamic therapy – a phased light effect on the wound surface with different wavelengths λ = 530, 630, 470 nm. Phagocytic activity of neutrophils using fluorescent label acridine orange (AO) – the ability of enzymes to cause denaturation of the double-stranded DNA of microorganisms inside phagocytes, the oxidative capacity of neutrophil enzymes in oxygen-dependent phagocytosis (NBT-test), the formation of extracellular neutrophilic traps (NETs), changes in the ratio of T lymphocyte subpopulations by expressing CD differentiation clusters (CD3+HLA-DR+, CD3+CD4+CD28+, CD3+CD4+CD25+CD127neg); lymphocyte ability to blast transformation according to the intensity of DNA synthesis of blast cells and concentration of IL-10 were determined. We used the methods of light and fluorescent microscopy (Olympus BX53), ELISA (Stat-Fax 3200) and flow cytometry (Cytomics FC500, Beckman Coulter).
