To highlight recent advances in NIR fluorophores, especially their rational design and application in NIR fluorescence imaging of the brain, including brain tumors, traumatic brain injuries and bleeding diseases, as well as neurodegenerative disorders like Alzheimer’s and Huntington’s diseases.

NIR-II imaging is a powerful tool for structural and functional characterization, among which the NIR-IIb imaging with a deeper tissue penetration is the most promising opportunity for brain imaging. Safe and effective fluorescent probes need to be continuously exploited to meet the clinical needs.
