PDPK-1 and other intrinsic kinases downstream of IGF-1 are key regulators of rod photoreceptor formation. Inhibition of PDPK-1 increases rod photoreceptor numbers, which depends on PKC activity. The study identifies a novel pathway involving P70-S6K and mTORC2 that stimulates rod development independently of PKC activity.

1:Experimental Design and Method Selection:
The study used a previously described ex vivo retinal explant cultured system to study photoreceptor development. Retinal explants were treated with various reagents to interrogate the effects of the IGF-1 pathway on photoreceptor development.

Mouse retinas were dissected from PN1 pups and cultured in a defined medium.

Retinal explants were cultured in UltraCulture medium supplemented with gentamicin. Western blots were performed using CytoBuster Protein Extraction Reagent, protease inhibitors, and phosphatase inhibitors.
4:Experimental Procedures and Operational Workflow:
Retinal explants were cultured for 96 h with or without reagents prior to fixation. Western blots were performed to analyze protein levels. Immunolabeling of retinal tissue was performed to study protein expression.
5:Data Analysis Methods:
Semi-quantitative analysis of band intensity was performed using ImageJ. Statistical analyses were performed using GraphPad Prism.
