The study demonstrates that raw Raman intensity increases with particle size up to a certain value, which depends on tablet width. Spectral preprocessing reduces the differences in Raman intensities due to particle size but does not completely eliminate it for the smallest particles. The preprocessing also corrects the mapping site-to-site differences in elastic scattering for tablets containing particles with predefined sizes. The findings are relevant for the quantitative application of Raman spectroscopy in pharmaceutical analysis.

The study is limited to the effect of particle size on Raman spectra of compacted samples of a single compound (KHP). The findings may not be directly applicable to complex mixtures or other materials. The spectral preprocessing reduces but does not completely eliminate the effect of particle size for the smallest particles (< 20 μm).
1:Experimental Design and Method Selection:
The study involved preparing tablets of potassium hydrogen phthalate (KHP) with different particle sizes to investigate the effect on Raman spectra. Two different instrumental optics were used: a macro-Raman system and a Raman microscope.
2:Sample Selection and Data Sources:
KHP was chosen for its purity, stability, and chemical structure. Tablets were prepared from KHP fractions of different particle sizes obtained by dry sieving.

A Brüker MultiRam spectrometer (macro-Raman system) and a RamanScope III (Raman microscope) were used. A Perkin Elmer

Spectra were recorded in two ways: repeatability (10 measurements at the same point) and mapping (10 measurements at different points). Spectral preprocessing included baseline correction and unit vector normalization.
