Maltol inhibits apoptosis of R28 cells induced by H2O2. The mechanisms underlying this protection are associated with NF-kB and MAPK-related pathways. Maltol could be an innovative neuroprotective therapeutic agent for the reduction of neurosensory retinal cell injury related to hypoxia.

R28 cells have both glial and neuronal characteristics, and do not behave like primary retinal ganglion cells (RGCs) in every aspect. Isolating and culturing primary RGCs are technically difficult.
1:Experimental Design and Method Selection:
R28 cells were exposed to hydrogen peroxide (H2O2) as an oxidative stress with or without maltol. Cell viability was monitored with the lactate dehydrogenase assay and apoptosis was examined by the TUNEL method. The expression and phosphorylation of NF-κB, ERK, JNK, and p38 were evaluated by Western immunoblot analysis.
2:Sample Selection and Data Sources:
R28 cells, which are rat embryonic precursor neuroretinal cells, were used.
3:List of Experimental Equipment and Materials:
Dulbecco’s modified Eagle’s medium, fetal bovine serum, streptomycin, hydrogen peroxide (H2O2), maltol, lactate dehydrogenase assay kit, TUNEL assay kit, Western immunoblot analysis equipment.
4:Experimental Procedures and Operational Workflow:
R28 cells were cultured and exposed to H2O2 and maltol. Cell viability and apoptosis were measured. The expression and phosphorylation of NF-κB, ERK, JNK, and p38 were evaluated.
5:Data Analysis Methods:
Statistical tests to determine differences between groups were performed with one-way analysis of variance, followed by the post hoc Tukey test using SPSS ver. 17.0.
