Ultrasensitive detection of diclofenac in water samples by a novel surface-enhanced Raman scattering (SERS)-based immunochromatographic assay using AgMBA@SiO2-Ab as immunoprobe
摘要: In the last years, surface-enhanced Raman scattering based immunochromatographic assay (SERS-ICA) has been getting great attention and been applied for the detection of different target analytes. In this study, a novel SERS-ICA using AgMBA@SiO2-Ab as an immunoprobe has been developed for rapid, quantitative and ultrasensitive detection of a pharmaceutical diclofenac (DCF) residue in water samples. Ag, Ag@SiO2 and AgMBA@SiO2 nanoparticles (NPs) were synthesized and characterized. The immunoprobe was prepared by sandwiching the Raman reporter mercaptobenzoic acid (MBA) between the core-shell layers and immobilizing the monoclonal antibody (mAb) against DCF on the surface of the Ag@SiO2 NPs. Under optimal conditions, the sensitivity and limit of detection (LOD) of the SERS-ICA for DCF were found to be 9 pg mL-1 and 0.07 pg mL-1, respectively. The detection of the SERS-ICA for DCF was completed within 15 min. AgMBA@SiO2-Ab was stable within four months without significant loss of SERS intensity. The key to success of the SERS-ICA was the employment of the Ag@SiO2 NPs as the SERS substrate, which possessed gorgeous SERS enhancement capability, predominant biocompatibility and excellent stability. This SERS-ICA is potential as an ultrasensitive analytical method in point-of-care testing, clinical diagnosis, food inspecting and environmental monitoring.

[Environmental Chemistry for a Sustainable World] Nanosensors for Environmental Applications Volume 43 || Development of Optical Sensor Strips for Point-of-Care Testing for Pesticide
摘要: Disposable or point-of-care sensors are a promising tool for low-cost and rapid sensing of analytes including pesticides. They find important applications in pesticide-contaminated food, agro-products, and water quality monitoring. This chapter highlights the implication and significance of pesticide residue identification in foodstuffs and overviews the most frequently engaged analytical techniques, and finally their benefits and limitations are discussed. Disposable strip-based biosensors have their intrinsic advantages and some disadvantages, but their cost-effectiveness and portability have turned them as a potential possibility for point-of-care (POC) testing of various pesticides. The fabrication of robust, low-cost, reliable, and sensitive sensors with the aid of both simple naked eye-based and portable readout-based detectors is the driving factor in this sensor’s technology area. The pending limitations can be overcome by adapting new specific recognition elements and better signal generative particles or systems. The integration of these devices with card readers or smartphones can make them more user-friendly and will provide more accurate quantitative information.

Time-resolved fluorescent immunochromatographic assay-based on three antibody labels for the simultaneous detection of aflatoxin B <sub/>1</sub> and zearalenone in Chinese herbal medicines
摘要: A time-resolved fluorescent immunochromatographic assay (TRFICA) was successfully developed for the sensitive, simultaneous, and quantitative detection of aflatoxin B1 (AFB1) and zearalenone (ZEN) in Chinese herbal medicines. Eu-nanospheres (EuNPs) with unique optical properties increased the stability and sensitivity of the immunochromatographic assay. To obtain stable quantitative results, we applied a three-label system in which monoclonal antibodies for AFB1 and ZEN were conjugated to the EuNPs as detection probes on the test line (T line), and EuNP-labelled chicken IgY conjugates acted as the reference on the control line (C line). The fluorescence intensities of the T and C lines were recorded, and the T/C ratio was employed as the quantitative signal for the elimination of strip variation and matrix effects. The parameters that affected the TRFICA were optimised. Under optimal conditions, the established TRFICA gave good linear ranges from 0.60 μg/kg to 3.92 μg/kg for AFB1 and from 0.40 μg/kg to 1.28 μg/kg for ZEN. The limits of detection for AFB1 and ZEN were as low as 0.60 and 0.40 μg/kg, respectively, in Chinese herbal medicines Semen coicis, Rhizoma dioscoreae, and Platycodon grandiflorus, respectively. The average recoveries of the spiked samples were 73%–95% for AFB1 and 75.83%–90% for ZEN, both with a relative standard deviation of < 9.08%. The results of 15 actual samples detected by the developed TRFICA showed a satisfactory correlation with those of ultra-performance liquid chromatography tandem mass spectrometry. Therefore, the TRFICA is a simple, rapid, and sensitive approach to quantitatively detect mycotoxins in Chinese herbal medicines.

Immunochromatographic assay for melamine based on luminescent quantum dot beads as signaling probes
摘要: To screen and detect the harmful substance melamine (MEL), a quantum-dot-bead-based immunochromatographic assay (QB-ICA) was formulated. After optimization, calibration was performed within the linear range from 0.06 to 0.28 ng mL?1, with limit of detection (LOD) of 0.04 ng mL?1. The LOD was 35 times lower than that of ICA that used colloidal gold nanoparticles (LOD ? 1.4 ng mL?1) and 40 times lower than that of the assay based on quantum dots (LOD ? 1.6 ng mL?1). In the detection of MEL in spiked pure milk using the proposed QB-ICA strategy, the LOD (LOD ? 0.19 ng mL?1) of the samples with the proposed pretreatment was 18.4 times lower than those of the samples without pretreatment (LOD ? 3.5 ng mL?1). The performance and practicability of the proposed QB-ICA system was validated; the obtained results reveal that QB-ICA is comparable with the conventional enzyme-linked immunosorbent assay (ELISA) method, but with enhanced applicability. Given its high sensitivity and practicability, the QB-ICA strategy could become a worthwhile alternative for the rapid, sensitive, and quantitative onsite detection of harmful substances, facilitating food safety monitoring.

Immunochromatographic assay for T-2 toxin based on luminescent quantum dot beads
摘要: A quantum dot bead based immunochromatographic assay (QB-ICA) system was established for T-2 toxin (T-2), which widely occurs in agriculture and could be used as a potential biological warfare agent. After optimization, the dynamic linear detection range of T-2 calculated from a calibration curve was from 0.12 to 0.67 ng mL(cid:1)1 and the limit of detection (LOD) was 0.08 ng mL(cid:1)1, which is lower than those of the ICA based on colloidal gold nanoparticles or a ?uorescent material or an antibody-based biochip in other reports. The performance and practicability of the established ICA system were validated with a commercial ELISA kit and the two methods were comparable. The proposed QB-ICA for T-2 could be an alternative for rapid, sensitive, and quantitative on-site detection of this toxin in biosafety monitoring in agriculture and for susceptibility testing of the potential release of this biological warfare agent.

Rapid immunological detection of copper ions using fluorescence immunochromatographic assay
摘要: One-step fluorescence immunochromatographic assay (FICA) has been developed using fluorescent microspheres-labelled monoclonal antibody (McAb) probe for the rapid detection of copper ions in herbal plants and soil samples were investigated in this study. The results can be read by the fluorescence reader according to the fitted standard curve equation. Under optimal conditions, detecting copper ions with FICA can be done within 15 min with a LOD (limit of detection) of 0.2 μg/L, and was very useful for the rapid onsite testing. A total of 31 solid samples were pre-treated by microwave digestion combined with cation exchange solid phase extraction and prepared as test samples for FICA and ICP-AES. The two methods showed good relativity with correlation coefficients of 0.924 and 0.969 in herbal plants and soil samples for copper detection, respectively. We hope that our research can help promote the daily monitoring of heavy metal pollution effectively.
