  • New Magneto-Fluorescent Hybrid Polymer Nanogel for Theranostic Applications

    摘要: Herein we have reported a new magneto-fluorescent nanogel built on photoluminescent co-macromer [PEG-maleic acid-glycine], N, N Dimethyl amino ethyl methacrylate and citrate capped superparamagnetic iron oxide nanoparticles (SPION). The nanogel was found to have core-shell morphology (SPION core and PEG shell) with particle size around 80 nm. The cytocompatibility of the synthesized nanogel studied using MTT, live dead assays and flow cytometry. The cellular uptake of the nanogel on cervical cancer cell line Hela evaluated through Prussian blue staining and fluorescence microscopy have revealed good cancer cell imaging capability. Magnetic hyperthermia experiments have shown that the synthesized nanogel caused the lysis of cancer cells. The fluorescence bio-imaging capability of the nanogel in murine model has shown good near IR imaging capability. Overall, the reported results suggest that the magneto-fluorescent nanogel shows promising future potential for cancer theranostic applications.

  • Fast responsive photo-switchable dual-color fluorescent cyclodextrin nanogels for cancer cell imaging

    摘要: Photo-switchable dual-color fluorescent nanogels were fabricated via nanoprecipitation in aqueous solution. The spiropyran-modified β-cyclodextrin (β-CD-SP) and 4-amino-7-nitro-1,2,3-benzoxadiazole (NBDNH2) were encapsulated into nanogels in the presence of 1,6-hexamethylene diisocyanate (HMDI). In the nanogels, spiropyran moiety acted as fluorescence molecule acceptors under the irradiation of UV or visible light to quench or recover the fluorescence of the NBDNH2 to achieve reversible dual-color fluorescence. The feed ratio of the two fluorophores played vital role on the energy transfer efficiency (E) of the nanogels, and the results showed that high E (90.7%) can be achieved when the feed ratio of NBDNH2/β-CD-SP is 1:2 (mol/mol). In addition, the nanogels can maintain good photo-switchable fluorescent behavior as long as 5 weeks. With rapid photo-responsiveness (within 60 s), good reversibility, long-term stability, and excellent cytocompatibility, the as-prepared nanogels have been successfully applied to photo-switchable dual-color fluorescent imaging in cancer cell.

  • Magnetic iron oxide nanoparticles/10-hydroxy camptothecin co-loaded nanogel for enhanced photothermal-chemo therapy

    摘要: Iron oxide nanoparticle is a promising nano-biomaterial for treating iron deficiency, or as drug carriers and contrast agent as well as stimulator to elicit ROS generation. However, because of their intrinsic structure, therapeutic agents are hard to be loaded by them alone. Nanogel is a promising candidate for co-loading of iron oxide and therapeutic drug, due to its network structure and permeability. In this study, we use pre-prepared nanogel as surfactant to co-load hydrophobic magnetic nanoparticles (MNPs) and anti-cancer drug (10-hydroxy camptothecin, HCPT) to construct MNPs/HCPT-nanogel for cancer diagnosis and therapy. By in vitro and in vivo evaluation, we reveal that the obtained MNPs/HCPT-nanogel are stable in aqueous environment and can be served as drug delivery system to efficiently inhibit the tumor growth as well as MRI contrast agent for MR imaging. The introduction of MNPs elicits the generation of ROS with the presence of macrophage, which can further inhibit the growth of cancer cell. Furthermore, the introduction of extra-magnetic field can further enhance the enrichment of MNPs/HCPT-nanogel in the tumor site, which further improves the chemotherapeutic outcome. Besides, the obtained MNP/HCPT-nanogel can be used as nanocarrier for photothermal therapy for its absorption at NIR region. With the combination of PTT and chemotherapy, not only can the growth primary tumor be inhibited, but also the metastasis can be alleviated. It demonstrates that the MNP/HCPT-nanogel can be served as a promising candidate for cancer PTT/chemotherapy to inhibit tumor growth and alleviate metastasis.

  • Facile Synthesis of Resveratrol Nanogels with Enhanced Fluorescent Emission

    摘要: Herein, a kind of fluorescent resveratrol nanogels via one-pot thiol-ene Michael addition polymerization of resveratrol triacrylate, 1,6-hexanedithiol, and methoxyl poly(ethylene glycol) acrylate is prepared. The resultant nanogels can be well-dispersed in water with a hydrodynamic radius of around 68 nm, and the nanogels are stable in both water and organic solvents. Moreover, the resveratrol nanogels exhibit elevated fluorescence intensity compared to free resveratrol, and the quantum yield of resveratrol nanogels is estimated to be 5.8 times as that of free resveratrol dispersed in water. Fluorescence image results also demonstrate that the resveratrol nanogels can be used for cell imaging in MCF-7 human breast cancer cells. Therefore, the resveratrol nanogels are expected to be used as a trackable drug delivery system.
