  • Systematic Truncating Aptamers to Create High Performance Graphene Oxide (GO)-based Aptasensors for Multiplex Detection of Mycotoxins

    摘要: Graphene Oxide (GO)-based aptasensor is currently one of the most popular sensing platforms for simple and rapid detection of various targets. Unfortunately, the GO-based aptasensors with long aptamer strands typically show unsatisfactory performance resulted from insignificant structural transformations upon target bindings. We report herein the utilization of an aptamer truncating strategy to combat such a challenge. Taking a pre-selected anti-aflatoxin B1 (AFB1) aptamer (P-AFB1-50) as a trial system, we sequentially remove the extraneous nucleotides within the aptamer by means of circular dichroism (CD) spectroscopy and binding affinity analysis. Particularly, the ratio of the quenching constants between the GO sheets and the truncated aptamers (labelled with fluorophores) in the absence and presence of target was determined for each of the truncated aptamers to evaluate the optimal sequence. As a result, the truncated aptamer comprising 40 nucleotides was confirmed to show the highest FL output and best detection limit upon conjugation with GO sheets. More importantly, we demonstrated that this truncating strategy is versatile, i.e., it can be easily extended to other aptamer systems (anti-ochratoxin A (OTA) aptamer, P-OTA-61, as an example) for extraneous nucleotide identification. Impressively, the two optimal truncated aptamers can work together on GO sheets to achieve a simultaneous detection of two different mycotoxins (i.e., AFB1 and OTA) in one single testing. Essentially, this research opens a new avenue for the design and testing of aptamer/GO based-sensing platforms for rapid, low-cost and multiplex quantification of analytical targets of interest.

    摘要: Analog filters for biomedical signal processing applications deals with very slow or low frequency electrical activities of the physiological signals. This paper proposes first order, second order, fifth order elliptic low pass, second order notch and high pass OTA-C filters using hybrid CMOS-CNFET technology. Carbon Nanotube Field Effect Transistors (CNFETs) and CMOS devises can be heterogeneously integrated on a single 3-D chip to realize important signal processing building blocks such as OTA-C filters. Proposed circuits use Operational Transconductance Amplifier (OTA) as a building block for OTA-C filters. Realized filter circuits satisfy ultra-low power consumption requirement of wearable and implantable biomedical devices. The transistors used in the circuit operate in weak inversion to achieve ultra-low power consumption.

  • Successful and quick treatment of nevus of Ota with 755nm picosecond laser in Chinese

    摘要: Nevus of Ota, also known as nevus fusco-caeruleus ophthalmo-maxillaris, is a benign dermal melanocytosis. In the past, this disease was usually treated by Q-switched laser therapy, but the course of treatment was relatively long. In recent years, it has been reported that 755nm picosecond laser, which was firstly reported to treat tattoos, is also effective in the treatment of nevus of Ota. Here, we report six cases of nevus of Ota which were treated with 755nm picosecond laser in Chinese people. We find amazingly that these lesions almost disappeared after only one or two sessions of treatment.

    摘要: The characterization of the performance of wireless devices is key to developing new RF products conforming to the latest communications protocols. Traditionally, communication performance tests have focused on the RF performance of the tested devices, e.g., smart phones, pads, laptops, and so on. In particular, the focus has shifted from conducted (i.e., cabled) measurements to more realistic over-the-air (OTA) characterization of the RF performance of these devices in transmit or receive mode. For example, the receiver performance of 2G and 3G wireless devices can be measured in terms of the total isotropic sensitivity (TIS) that depends on the antenna and the receiver parts of a wireless device. These measurements can be performed in a reverberation chamber setup. However, standard TIS measurements can be time-consuming and do not reflect the actual performance gains of multiple-input multiple-output antenna systems operating over orthogonal frequency division multiplexing channels, such as those encountered in 4G long-term evolution (LTE) systems. Therefore, in order to meet both time and cost efficiency requirements, we propose here a new method to determine the TIS, as well as the diversity performance, of an LTE device based on throughput measurements. The proposed method shows that the TIS of an LTE device is characterized much faster directly from OTA throughput measurements than from standard TIS measurements and with excellent accuracy.

  • Use of a Picosecond Alexandrite Laser for Treating Acquired Bilateral Nevus of Ota-Like Macules in Chinese Patients

    摘要: Background and Objectives: Acquired bilateral nevus of Ota‐like macules (ABNOM) is common among the Chinese population. There have been few studies on the use of a picosecond alexandrite laser for the treatment of ABNOMs. The goal of this study was to evaluate the efficacy and safety of a 755 nm picosecond alexandrite laser for the treatment of ABNOMs. Study Design/Materials and Methods: A total of 225 subjects diagnosed with ABNOMs were included in this retrospective study. A 755 nm picosecond alexandrite laser system was used to treat all subjects. The safety and effectiveness of the laser were determined using a clinical photo assessment. The correlating factors of the patients, including the age, Fitzpatrick skin type, lesion location, lesion color, treatment procedures, melasma, and outcomes, were analyzed. Results: The patients underwent one to four treatment sessions with clinical clearance (>90%) rates of 8.89%, 30.99%, 56.65%, and 60.00%, respectively. A higher number of sessions was positively associated with a better treatment response. The hyperpigmentation incidence rate was 22.67%, and one case of hypopigmentation was observed. Conclusion: Our results suggest that a picosecond alexandrite laser is an effective and safe approach for treating ABNOMs in Chinese patients.

  • Successful treatment of a patient with multicolored nevus of Ota using a combination of 1064 and 532 nm Q-switched Nd:YAG lasers

    摘要: Nevus of Ota (NO) is a hamartoma of dermal melanocytes usually presents as unilateral blue, gray or brown macules or patches. It can impose a high burden of cosmetic and psychological disturbances in affected individuals. Q-Switched lasers appear to be an effective treatment for this kind of dermal melanocytosis. Multicolored Ota nevus is a rare variant of this kind of nevus and its treatment may be more challenging compared with unicolor lesions. Herein we report a 21-year-old woman with a multicolored nevus of Ota (blue and brown), which showed a dramatic response to a combination of 1,064 nm and 532 nm Q-Switched Nd:YAG lasers. We also discuss different aspects of the Q-switched laser application of Ota nevus treatment. We also focus on laser combination therapy to treat the nevus of Ota.

  • Comparison of a picosecond alexandrite laser versus a Q-switched alexandrite laser for the treatment of nevus of Ota: A randomized, split-lesion, controlled trial

    摘要: Background: Novel picosecond lasers have been available for various pigmentary disorders. However, there are limited data directly comparing picosecond lasers and Q-switched lasers for treatment of nevus of Ota. Objective: To compare the efficacy and safety of a picosecond alexandrite laser (PSAL) compared with a Q-switched alexandrite laser (QSAL) for the treatment of nevus of Ota. Methods: Each lesion of the 56 participants enrolled was split into two parts and randomly assigned to either PASL or QSAL treatment arm. Each lesion was treated up to 6 sessions at a 12-week interval. The efficacy and safety were determined based on blinded visual evaluation and self-report at each follow-up visit. Results: PSAL arm achieved a significantly better clearance (4.53 vs 4.0) with fewer sessions (5.26 vs 5.87) and less severe pain (5.61 vs 6.40). Patients were more satisfied with PSAL than QSAL (4.5 vs 4.0). Occurrences of post inflammatory hyperpigmentation (26% vs 34%) and hypopigmentation (21% vs 47%) were also lower in PSAL than QSAL arm. Limitations: Lack of objective assessments and outcome measures. Conclusions: PSAL demonstrated better clinical results and fewer adverse events than QSAL for the treatment of nevus of Ota.

  • Aptamers Improving Fluorescence Anisotropy and Fluorescence Polarization Assays for Small Molecules

    摘要: Nucleic acid affinity probes, such as aptamers, have been combined with fluorescence anisotropy (FA) / fluorescence polarization (FP) technology for the development of a diverse range of assays. Formation of a complex between a small fluorescent molecule and its binding partner usually increases the overall size of the fluorescent molecule and decreases its rate of rotation, resulting in increases in fluorescence anisotropy/polarization. Structure-switching of the fluorescently labeled aptamers arising from target binding can also affect molecular volume, local rotation of the fluorophore, and/or fluorescence lifetime, causing changes in anisotropy/polarization. Incorporation of the unique adsorptive properties of single-stranded nucleic acid aptamers on nanomaterials, hybridization of aptamers with complementary sequences, and the amplifiable ability of nucleic acid aptamers have broadened the applications of fluorescence anisotropy assays and enhanced their sensitivity. This review focuses on aptamer-based fluorescence anisotropy assays for the detection of small molecules, such as therapeutic drugs, environmental contaminants, natural toxins, and metabolites.
