  • Simultaneous Investigation of Vascular and Retinal Pigment Epithelial Pathologies of Exudative Macular Diseases by Multifunctional Optical Coherence Tomography

    摘要: To investigate exudative macular disease, multifunctional optical coherence tomography (MF-OCT) using a 1-lm probe band was developed. The clinical utility of MF-OCT was examined in a descriptive case series. METHODS. Ten eyes of nine subjects with exudative macular disease, including one eye with age-related macular degeneration (AMD), one eye with idiopathic neovascular maculopathy, and eight eyes with polypoidal choroidal vasculopathy (PCV), were investigated. Areas of 6 3 6 mm2 around the pathologic region were scanned with 512 3 1024 depth scans in 6.6 seconds. Structural OCT, Doppler optical coherence angiography (OCA), and cumulative phase retardation images were obtained with a single measurement. Each MF-OCT image visualized the structure, vasculature, and birefringence. Degree of polarization uniformity values were also obtained for selective visualization of the retinal pigment epithelium (RPE). The MF-OCT images were compared with conventional ophthalmic images. RESULTS. Abnormal vasculatures were observed with Doppler OCA in all eyes, which presented high similarity to indocyanine green angiography in the midphase. The RPE and exudation in the pathologic regions were discriminated in one eye with AMD and ?ve of eight eyes with PCV. Cumulative phase retardation visualized ?brosis scars in two of the PCV cases. CONCLUSIONS. Multifunctional OCT revealed depth-resolved abnormal vasculatures, the integrity of the RPE and choroid, discrimination of the RPE and exudation, and existence of ?brosis scars in exudative macular diseases. Interpretation of MF-OCT examination is well matched with conventional ophthalmic examination. These results suggest that MF-OCT can be used as a noninvasive ophthalmic examination tool prior to conventional examinations in clinical routines.

  • Blood–Retinal Barrier Compromise and Endogenous <i>Staphylococcus aureus</i> Endophthalmitis

    摘要: PURPOSE. To test the hypothesis that blood–retinal barrier compromise is associated with the development of endogenous Staphylococcus aureus endophthalmitis. METHODS. To compromise the blood–retinal barrier in vivo, streptozotocin-induced diabetes was induced in C57BL/6J mice for 1, 3, or 5 months. Diabetic and age-matched nondiabetic mice were intravenously injected with 108 colony-forming units (cfu) of S. aureus, a common cause of endogenous endophthalmitis in diabetics. After 4 days post infection, electroretinography, histology, and bacterial counts were performed. Staphylococcus aureus–induced alterations in in vitro retinal pigment epithelial (RPE) cell barrier structure and function were assessed by anti–ZO-1 immunohistochemistry, FITC-dextran conjugate diffusion, and bacterial transmigration assays. RESULTS. We observed one bilateral infection in a control, nondiabetic animal (mean ? 1.54 3 103 6 1.78 3 102 cfu/eye, 7% incidence). Among the 1-month diabetic mice, we observed culture-con?rmed unilateral infections in two animals (mean ? 5.54 3 102 6 7.09 3 102 cfu/ eye, 12% incidence). Among the 3-month diabetic mice, infections were observed in 11 animals, three with bilateral infections (mean ? 2.67 3 102 6 2.49 3 102 cfu/eye, 58% incidence). Among the 5-month diabetic mice, we observed infections in ?ve animals (mean ? 7.88 3 102 6 1.08 3 103 cfu/eye, 33% incidence). In vitro, S. aureus infection reduced ZO-1 immunostaining and disrupted the barrier function of cultured RPE cells, resulting in diffusion of ?uorophore-conjugated dextrans and transmigration of live bacteria across a permeabilized RPE barrier. CONCLUSIONS. Taken together, these results indicated that S. aureus is capable of inducing blood–retinal barrier permeability and causing endogenous bacterial endophthalmitis in normal and diabetic animals.

  • Incorporation of K<sup>+</sup> and Al<sup>3+</sup> during the formation of Ta<sub>3</sub>N<sub>5</sub> with heat-treatment of Ta<sub>2</sub>O<sub>5</sub>–AlN mixtures without flowing ammonia

    摘要: A tantalum(V) nitride (Ta3N5)-containing powder for non-toxic red pigments was prepared by heat-treatment for the mixed powders of tantalum(V) oxide (Ta2O5), aluminum nitride and potassium fluoride in nitrogen atmosphere without flowing ammonia. The foreign ions of K+ and Al3+ were incorporated into Ta3N5 in the synthetic process. The incorporation of the ions had an influence on the hue of Ta3N5. It can coordinate a reddish color of Ta3N5 pigments.

  • Alexandrite laser induced uveitis & pigment dispersion: A case report and review of the literature

    摘要: To describe a complication of an upper eyelid alexandrite laser procedure. Observations: A 55-year-old woman presented with left eye blurred vision and photophobia after a left upper eyelid procedure with an alexandrite laser. She had elevated intraocular pressure (IOP), anterior chamber cell and pigment, posterior synechiae, and retroillumination defects in the left eye. She was treated with topical prednisolone and brimonidine. Six months later, although her anterior chamber had cleared and IOP had normalized, the patient reported decreased vision-related quality of life from persistent photophobia. Conclusions and Importance: Alexandrite lasers are commonly used for hair removal and skin depigmentation. When used periocularly without proper eye protection, they have the potential to create irreversible ocular complications. This case demonstrates the importance of proper eye protection with periocular laser procedures.

  • White Painting Pigment as a Low-Cost Light Scattering Material for Bilayer Photoelectrodes of Dye-Sensitized Solar Cells

    摘要: White pigment (DuPont R902+) has been used as a light scattering material in the preparation of bilayer photoelectrodes of dye-sensitized solar cells (DSCs). The X-ray diffraction (XRD) pattern of the white pigment revealed that the material consists of rutile phase of titanium dioxide. The light scattering layer prepared from the white pigment was coated onto the main-layer of the photoelectrodes of DSCs. The solar cells with and without light scattering layer were tested in the simulated light of 100 mW/cm2. The DSCs with the light scattering layer generated more current density than the DSCs without scattering layer and the overall light to electric power conversion efficiency of DSCs with the light scattering layer was ~4.00 % compared with 3.25 % efficiency of the DSCs without the scattering layer.

  • A simple, static preservation system for shipping retinal pigment epithelium cell sheets

    摘要: The ability to move cells and tissues from bench to bedside is an essential aspect of regenerative medicine. In this study, we propose a simple, static shipping system to deliver tissue-engineered cell sheets. Notably, this system is electronic-device-free and simplified to minimize the number of packing and opening steps involved. Shipping conditions were optimized, and application and verification of the system were performed using human iPS-cells-derived or fetal retinal pigment epithelium (RPE) cell sheets. The temperature of the compartments within the insulated container was stable at various conditions, and filling up the cell vessel with medium effectively prevented turbulence-induced mechanical damage to the RPE cell sheets. Furthermore, no abnormal changes were observed in RPE morphology, transepithelial electrical resistance, or mRNA expression after transit by train and car. Taken together, our simple shipping system has the potential to minimize the costs and human error associated with bench to bedside tissue transfer. This specially designed regenerative tissue shipping system, validated for use in this field, can be used without any special training. This study provides a procedure for easily sharing engineered tissues with the goal of promoting collaboration between laboratories and hospitals and enhancing patient care.

  • All-trans retinoic acid stimulates the secretion of TGF-β2 via the phospholipase C but not the adenylyl cyclase signaling pathway in retinal pigment epithelium cells

    摘要: Background: By investigating that (i) all-trans retinoic acid (ATRA) affects human retinal pigment epithelium (RPE) in expressing and secreting transforming growth factor (TGF)-β2 and (ii) U73122 (phospholipase C inhibitor) and SQ22536 (adenylyl cyclase inhibitor) regulate the ATRA-induced secretion of TGF-β2 in human RPE, we sought to interpret the signaling pathway of ATRA in promoting the development of myopia. Methods: The RPE cell line (D407) was treated with (i) ATRA (10 μM), (ii) U73122 (5–40 μM) and ATRA (10 μM), or (iii) SQ22536 (5–40 μM) and ATRA (10 μM). The control group was no-treated. After stimulated at 2, 4, 8, 16, 24, and 48 h, The expression and secretion of TGF-β2 was detected. Results: TGF-β2 in the cytoplasm was time-dependent increased by ATRA (p < 0.001). A time-dependent increase in the TGF-β2 protein of the supernatant was induced by ATRA (p < 0.001). U73122 (in the range of 5 to 40 μM) could suppress the secretion of TGF-β2 induced by ATRA (p < 0.001), and 40 μM U73122 could completely inhibit the up-regulated effect of 10 μM ATRA. However, SQ22536 (in the range of 5 to 40 μM) had no impact on the secretion of TGF-β2 induced by ATRA (p > 0.05). Conclusions: In RPE cells, ATRA stimulates the secretion of TGF-β2 via the phospholipase C signaling pathway but not the adenylyl cyclase signaling pathway. U73122 may inhibit the promotion of ATRA in the development of myopia.

  • Prognostic phenotypic and genotypic factors associated with photodynamic therapy response in patients with age-related macular degeneration

    摘要: Background: This study aimed to demonstrate the phenotypic and genotypic factors associated with photodynamic therapy (PDT) for age-related macular degeneration (AMD). Methods: The study included 149 patients with exudative AMD treated by PDT. Eight phenotypic factors and ten genotypic factors for three single nucleotide polymorphisms (SNPs; rs800292, rs1061170, rs1410996) in the complement factor H (CFH) gene, rs 11200638-SNP in the high temperature requirement A-1 (HTRA1) gene, two SNPs (rs699947, rs2010963) in the vascular endothelial growth factor (VEGF) gene, and four SNPs (rs12948385, rs12150053, rs9913583, rs1136287) in the pigment epithelium-derived factor (PEDF) gene were evaluated. Results: A significant association with best-corrected visual acuity change was demonstrated in the greatest linear dimension, presence or absence of pigment epithelial detachment, and HTRA1-rs11200638 genotype statistically (P=3.67×10-4, 1.95×10-2, 1.24×10-3, respectively). Best-corrected visual acuity in patients with AA genotype of HTRA1-rs11200638 significantly decreased compared with that in patients with GG genotype (P=1.33×10-3). Logistic regression analyses demonstrated HTRA1-rs11200638 genotype was most strongly associated with best-corrected visual acuity outcome from baseline at 12 months after photodynamic therapy (P=4.60×10-3; odds ratio 2.363; 95% confidence interval 1.303–4.285). Conclusion: The HTRA1-rs11200638 variant showed the most significant association. Therefore, this variant may be used as a prognostic factor to estimate the PDT response with significant predictive power.

  • The Study on Titanium Dioxide-Silica Binary Mixture Coated SrAl2O4: Eu2+, Dy3+ Phosphor as a Photoluminescence Pigment in a Waterborne Paint

    摘要: After proper stimulation, long afterglow phosphors formulated as pigment in waterborne paints can emit light after the removal of the excitation light source. The encapsulation of SrAl2O4: Eu2+, Dy3+ phosphor by TiO2 and SiO2 individually, and in combination by a precipitation method was studied. The water resistance and photoluminescence behavior of the coated phosphors as a pigment for potential use in waterborne photoluminescence paints were evaluated. It revealed that the TiO2- SiO2 content coating layer was precipitated on the studied phosphor successfully. The higher trend of the TiO2 for coating on the phosphor when compared with the SiO2 was observed from EDS spectra. The SEM micrographs showed a continuous and uniform SiO2-TiO2 layer on the coated phosphor. Based on XRD results, the existence of the TiO2 in the coating layer had beneficial effect on the average crystallite size values. The pH solution versus time showed that the availability of the TiO2 in coated layer improved water resistance of the coated phosphor, although, in comparison with SiO2, it was less effective. The coated phosphor with TiO2 had the minimum afterglow brightness decay and consequently, it was recommended as a suitable pigment for waterborne photoluminescence paints.

  • Inhibitor of growth 4 affects hypoxia-induced migration and angiogenesis regulation in retinal pigment epithelial cells

    摘要: Inhibitor of growth 4 (ING4), a potential tumor suppressor, is implicated in cell migration and angiogenesis. However, its effects on diabetic retinopathy (DR) have not been elucidated. In this study, we aimed to evaluate ING4 expression in normal and diabetic rats and clarify its effects on hypoxia‐induced dysfunction in human retinal pigment epithelial (ARPE‐19) cells. A Type 1 diabetic model was generated by injecting rats intraperitoneally with streptozotocin and then killed them 4, 8, or 12 weeks later. ING4 expression in retinal tissue was detected using western blot analysis, reverse transcription quantitative real‐time polymerase chain reaction (RT‐qPCR), and immunohistochemistry assays. After transfection with an ING4 overexpression lentiviral vector or small interfering RNA (siRNA), ARPE‐19 migration under hypoxia was tested using wound healing and transwell assays. The angiogenic effect of conditioned medium (CM) from ARPE‐19 cells was examined by assessing human retinal endothelial cell (HREC) capillary tube formation. Additionally, western blot analysis and RT‐qPCR were performed to investigate the signaling pathways in which ING4, specificity protein 1 (Sp1), matrix metalloproteinase 2 (MMP‐2), MMP‐9, and vascular endothelial growth factor A (VEGF‐A) were involved. Here, we found that ING4 expression was significantly reduced in the diabetic rats’ retinal tissue. Silencing ING4 aggravated hypoxia‐induced ARPE‐19 cell migration. CM collected from ING4 siRNA‐transfected ARPE‐19 cells under hypoxia promoted HREC angiogenesis. These effects were reversed by ING4 overexpression. Furthermore, ING4 suppressed MMP‐2, MMP‐9, and VEGF‐A expression in an Sp1‐dependent manner in hypoxia‐conditioned ARPE‐19 cells. Overall, our results provide valuable mechanistic insights into the protective effects of ING4 on hypoxia‐induced migration and angiogenesis regulation in ARPE‐19 cells. Restoring ING4 may be a novel strategy for treating DR.
