Spatio-temporal regulation of Rx and mitotic patterns shape the eye-cup of the photoreceptor cells in Ciona
摘要: The ascidian larva has a pigmented ocellus comprised of a cup-shaped array of approximately 30 photoreceptor cells, a pigment cell, and three lens cells. Morphological, physiological and molecular evidence has suggested evolutionary kinship between the ascidian larval photoreceptors and vertebrate retinal and/or pineal photoreceptors. Rx, an essential factor for vertebrate photoreceptor development, has also been suggested to be involved in the development of the ascidian photoreceptor cells, but a recent revision of the photoreceptor cell lineage raised a crucial discrepancy between the reported expression patterns of Rx and the cell lineage. Here, we report spatio-temporal expression patterns of Rx at single-cell resolution along with mitotic patterns up to the final division of the photoreceptor-lineage cells in Ciona. The expression of Rx commences in non-photoreceptor a-lineage cells on the right side of the anterior sensory vesicle at the early tailbud stage. At the mid tailbud stage, Rx begins to be expressed in the A-lineage photoreceptor cell progenitors located on the right side of the posterior sensory vesicle. Thus, Rx is specifically but not exclusively expressed in the photoreceptor-lineage cells in the ascidian embryo. Two cis-regulatory modules are shown to be important for the photoreceptor-lineage expression of Rx. The cell division patterns of the photoreceptor-lineage cells rationally explain the generation of the cup-shaped structure of the pigmented ocellus. The present findings demonstrate the complete cell lineage of the ocellus photoreceptor cells and provide a framework elucidating the molecular and cellular mechanisms of photoreceptor development in Ciona.

Theoretical Analysis of Moving Reference Planes Associated with Unit Cells of Nonreciprocal Lossy Periodic Transmission-Line Structures
摘要: This paper presents a theoretical analysis of moving reference planes associated with unit cells of non-reciprocal lossy periodic transmission-line structures (NRLSPTLSs) by the equivalent bi-characteristic-impedance transmission line (BCITL) model. Applying the BCITL theory, only the equivalent BCITL parameters (characteristic impedances for waves propagating in forward and reverse directions and associated complex propagation constants) are of interest. An infinite NRLSPTLS is considered first by shifting a reference position of unit cells along TLs of interest. Then, a semi-infinite terminated NRLSPTLS is investigated in terms of associated load reflection coefficients. It is found that the equivalent BCITL characteristic impedances of the original and shifted unit cells are mathematically related by the bilinear transformation. In addition, the associated load reflection coefficients of both unit cells are mathematically related by the bilinear transformation. However, the equivalent BCITL complex propagation constants remain unchanged. Numerical results are provided to show the validity of the proposed theoretical analysis.

Budget impact from the incorporation of positron emission tomography – computed tomography for staging lung cancers
摘要: OBJECTIVE: To estimate the budget impact from the incorporation of positron emission tomography (PET) in mediastinal and distant staging of non-small cell lung cancer. METHODS: The estimates were calculated by the epidemiological method for years 2014 to 2018. Nation-wide data were used about the incidence; data on distribution of the disease′s prevalence and on the technologies’ accuracy were from the literature; data regarding involved costs were taken from a micro-costing study and from Brazilian Unified Health System (SUS) database. Two strategies for using PET were analyzed: the offer to all newly-diagnosed patients, and the restricted offer to the ones who had negative results in previous computed tomography (CT) exams. Univariate and extreme scenarios sensitivity analyses were conducted to evaluate the influence from sources of uncertainties in the parameters used. RESULTS: The incorporation of PET-CT in SUS would imply the need for additional resources of 158.1 BRL (98.2 USD) million for the restricted offer and 202.7 BRL (125.9 USD) million for the inclusive offer in five years, with a difference of 44.6 BRL (27.7 USD) million between the two offer strategies within that period. In absolute terms, the total budget impact from its incorporation in SUS, in five years, would be 555 BRL (345 USD) and 600 BRL (372.8 USD) million, respectively. The costs from the PET-CT procedure were the most influential parameter in the results. In the most optimistic scenario, the additional budget impact would be reduced to 86.9 BRL (54 USD) and 103.8 BRL (64.5 USD) million, considering PET-CT for negative CT and PET-CT for all, respectively. CONCLUSIONS: The incorporation of PET in the clinical staging of non-small cell lung cancer seems to be financially feasible considering the high budget of the Brazilian Ministry of Health. The potential reduction in the number of unnecessary surgeries may cause the available resources to be more efficiently allocated.

A Multi-Well Thin-Si LAPS and All-in-One Readout System for Ion Activity Monitor of Epithelium Cells
摘要: To measure the ion activities of cells, an easy-access and fully-integrated system is necessary in culture room with high cleanness and easy maintenance. A new sensor structure integrated with readout system based on light-addressable potentiometric sensor (LAPS) to quantitatively monitor real-time cell activity with the advantages of label free and 2D image ability is proposed. The difference of cell number and acidification could be easily observed by 2D images by means of this proposed methodology.

The use of F-FDG PET/CT in testicular cancer
摘要: Testicular germ cell tumors are one of the most common neoplasms in young men. After inguinal orchiectomy is performed, treatment options range from active surveillance to chemotherapy, radiation therapy or retroperitoneal lymphadenectomy. For the accurate use of the different treatment options, precise imaging techniques are necessary to reduce side effects of the aggressive therapies but also to avoid undertreatment exposing the patient to the risk of recurrence. F-FDG PET/CT is already recommended in the European guidelines for the follow up of seminomas under certain conditions but conventional primary staging or restaging is still performed with CT or MRI. Recently, the importance of F-FDG PET/CT in this context has become an interesting topic of academic discussion and subject of various clinical trials which are reviewed in the following.

Novel Four-Port DC-DC Converter for Interfacing Solar PV-Fuel Cell Hybrid Sources with Low-Voltage Bipolar DC Microgrids
摘要: Bipolarity in dc microgrids is desirable as it enhances the system reliability and efficiency. However, a bipolar dc microgrid (BDCMG) demands multiple conventional dc-dc converters to feed power to both the poles of the bipolar dc grid. To handle this requirement and to maintain high efficiency, a new four-port, dual-input-dual-output (DIDO) converter topology is proposed for interfacing the solar PV and fuel cell (FC) sources to a low-voltage BDCMG. The proposed topology is unidirectional, efficient, and compact. It has fewer circuit elements with only one inductor as compared to the conventional non-isolated dc-dc converters. The proposed converter regulates one of the pole voltages of the dc bus and also ensures MPPT of the PV source. Further, the converter can be operated as a single-input-dual-output (SIDO) converter. The control complexity of the proposed converter is low as it can be operated in various modes with only one set of controllers. To design the control system for the proposed converter, a small-signal model is derived for each operating mode. Loss modelling and efficiency analysis of the proposed converter are carried out and its efficacy and performance are validated by detailed simulation and experimental results under various operating conditions.

Rapid Deposition of Photocatalytically Enhanced TiO<sub>2</sub> Film by Atmospheric SPPS Using Ar/N<sub>2</sub>-Vortex Plasma Jet
摘要: In this study, as cost effective and an environmentally friendly film deposition technology, Atmospheric Solution Precursor Plasma Spray (ASPPS) was utilized for the deposition of the photo-catalytic titanium oxide (TiO2) film for the fabrication process of photovoltaic devices for rural electrification in developing countries. In addition, ethanol-diluted titanium tetra-iso-butoxide (TTIB: Ti(OC4H9)4) was used as a feedstock. N2-dominant Ar/N2 was also utilized as plasma working gas as well as for the elevation of the thermal plasma energy. By controlling deposition distances and temperature, using vortex generation anode nozzle operated at 1 kW, photo-catalytic TiO2 film was deposited and its crystallinity was confirmed by X-ray diffraction. Besides, the photo-catalytic properties of the film were confirmed by the methylene blue decolorization and the surface wettability tests. Surface morphologies of the TiO2 film was evaluated using optical micrographs. Furthermore, the film thickness and strength were measured using micro screw gauge and pencil scratch tester respectively. Lastly, when this Photo-catalytic TiO2 film was applied to photovoltaic devices, the device generated an open circuit voltage of 146.7 mV with solar irradiance intensity of 574 W/m2. From these results it was confirmed that, the ASPPS technology equipped with high cooling efficiency vortex anode nozzle is available for deposition of TiO2 film for the fabrication process of low-cost photovoltaic devices for rural areas in developing countries.

MEMS-Based Thermoelectric-Photoelectric Integrated Power Generator
摘要: This letter presents a novel MEMS-based thermoelectric–photoelectric power generator, which integrates a micro-scale thermoelectric generator (μ-TEG) and a solar cell on a single silicon chip by MEMS technology for the first time. To optimize the heat flux path of μ-TEG, one side of the thermocouple square array is on the interdigitated electrode of the solar cell and the other side is on the thick oxide passivation layer to realize thermal isolation. Moreover, the hot side and the cold side are insulated by a thick polyimide thermal insulating layer above the thermopile, and a series of square holes is created to enhance the thermal coupling between the thermopile and surrounding environment. Several common measures, like the back-surface field and oxide passivation layer with limited contact openings, are adopted for the design of the solar cell. All the measuring electrodes are on the same side of the device for the convenience of bonding wire and package. When a sputtered Al membrane covers the front side of the device, the maximum output voltage factor and the power factor of the μ-TEG are 0.149 V · cm?2 · K?1 and 3.03 × 10?3μW · cm?2 · K?2, respectively. When the front and back sides of device receive light, the measured photoelectric conversion efficiencies are 4.45% and 0.682%, respectively.

Photochemically Active Dyes for Super-Resolution Microscopy
摘要: The development of super-resolved optical microscopies has revolutionized the way we visualize cell biology. These techniques strongly rely on the use of photochemically active fluorophores that display changes in their photophysical properties upon irradiation with light. Many reversible and irreversible photochemical transformations have been explored for this purpose, and different imaging techniques require specific mechanisms of photoconversion. In this review, we provide an overview of the most common strategies used for the development of fluorophores for super-resolution microscopy and give specific examples of state-of-the-art fluorogenic probes. Furthermore, we discuss their main field of application and possible directions for future developments.

Fluorescently Tagged CCL19 and CCL21 to Monitor CCR7 and ACKR4 Functions
摘要: Chemokines are essential guidance cues orchestrating cell migration in health and disease. Cognate chemokine receptors sense chemokine gradients over short distances to coordinate directional cell locomotion. The chemokines CCL19 and CCL21 are essential for recruiting CCR7-expressing dendritic cells bearing pathogen-derived antigens and lymphocytes to lymph nodes, where the two cell types meet to launch an adaptive immune response against the invading pathogen. CCR7-expressing cancer cells are also recruited by CCL19 and CCL21 to metastasize in lymphoid organs. In contrast, atypical chemokine receptors (ACKRs) do not transmit signals required for cell locomotion but scavenge chemokines. ACKR4 is crucial for internalizing and degrading CCL19 and CCL21 to establish local gradients, which are sensed by CCR7-expressing cells. Here, we describe the production of fluorescently tagged chemokines by fusing CCL19 and CCL21 to monomeric red fluorescent protein (mRFP). We show that purified CCL19-mRFP and CCL21-mRFP are versatile and powerful tools to study CCR7 and ACKR4 functions, such as receptor trafficking and chemokine scavenging, in a spatiotemporal fashion. We demonstrate that fluorescently tagged CCL19 and CCL21 permit the visualization and quantification of chemokine gradients in real time, while CCR7-expressing leukocytes and cancer cells sense the guidance cues and migrate along the chemokine gradients.
