Deep Learning in Image Cytometry: A Review
摘要: Artificial intelligence, deep convolutional neural networks, and deep learning are all niche terms that are increasingly appearing in scientific presentations as well as in the general media. In this review, we focus on deep learning and how it is applied to microscopy image data of cells and tissue samples. Starting with an analogy to neuroscience, we aim to give the reader an overview of the key concepts of neural networks, and an understanding of how deep learning differs from more classical approaches for extracting information from image data. We aim to increase the understanding of these methods, while highlighting considerations regarding input data requirements, computational resources, challenges, and limitations. We do not provide a full manual for applying these methods to your own data, but rather review previously published articles on deep learning in image cytometry, and guide the readers toward further reading on specific networks and methods, including new methods not yet applied to cytometry data.

Neurodegeneration in ocular and central nervous systems: optical coherence tomography study in normal-tension glaucoma and Alzheimer disease*
摘要: To evaluate, in vivo, the optical coherence tomography (OCT) of the retinal nerve fiber layer (RNFL) and ganglion cell complex (GCC) in patients with normal-tension glaucoma (NTG) and those with Alzheimer disease (AD) in comparison with healthy subjects. This cross-sectional study included 18 patients with NTG, 20 with AD, and 20 control subjects. An ophthalmologic examination and OCT scans of both eyes were performed in all patients. There was a significant reduction in peripapillary RNFL thickness and macular GCC thickness and a significant increase in the global loss volume (GLV) rate in both the NTG and AD patients when compared to the control subjects (P = 0.004, P = 0.006, P < 0.001, respectively). The statistical evaluation showed no difference in any RNFL or GCC parameters between the AD and NTG groups (P > 0.05). There was a negative correlation between disease duration and average RNFL and GCC thicknesses (r = ?0.350, P = 0.027 and r = ?0.471, P = 0.002, respectively) and a positive correlation between duration and GLV (r = 0.427, P = 0.006) in the AD group. The average RNFL thickness, GCC thickness, and GLV rates may help in the diagnosis of AD as an additional examination and may provide some important clues about the duration of the disease.

Label‐Free Metabolic Classification of Single Cells in Droplets Using the Phasor Approach to Fluorescence Lifetime Imaging Microscopy
摘要: Characterization of single cell metabolism is imperative for understanding subcellular functional and biochemical changes associated with healthy tissue development and the progression of numerous diseases. However, single-cell analysis often requires the use of ?uorescent tags and cell lysis followed by genomic pro?ling to identify the cellular heterogeneity. Identifying individual cells in a noninvasive and label-free manner is crucial for the detection of energy metabolism which will discriminate cell types and most importantly critical for maintaining cell viability for further analysis. Here, we have developed a robust assay using the droplet micro?uidic technology together with the phasor approach to ?uorescence lifetime imaging microscopy to study cell heterogeneity within and among the leukemia cell lines (K-562 and Jurkat). We have extended these techniques to characterize metabolic differences between proliferating and quiescent cells—a critical step toward label-free single cancer cell dormancy research. The result suggests a droplet-based noninvasive and label-free method to distinguish individual cells based on their metabolic states, which could be used as an upstream phenotypic platform to correlate with genomic statistics.

Computed tomography imaging characteristics of primary atrial intimal sarcoma
摘要: We present a 36-year-old man who presented to our emergency department with acute onset shortness of breath and syncope. He was found to have a large left atrial mass on initial computed tomography (CT) which was confirmed by echocardiography. Tumor biopsy and attempted excision were performed, showing a primary cardiac spindle cell sarcoma that was unable to be resected, to which the patient eventually succumbed to. Spindle cell sarcomas of the heart are very rare primary cardiac tumors, with a variable, non-specific presentation. The most effective treatment is surgical resection, with chemotherapy and radiotherapy showing some benefit. Despite these treatments, the prognosis is poor. Given the uncommon nature of this tumor, the objective of this report is to demonstrate the clinical presentation and CT imaging characteristics of a case of primary cardiac spindle cell sarcoma, to raise awareness of this entity, and to increase the index of suspicion as a potential differential diagnosis to cardiac tumors seen on imaging.

A ratiometric fluorescent probe for hydrazine detection with large fluorescence change ratio and its application for fluorescence imaging in living cells
摘要: Hydrazine and its derivatives are one of the most important raw chemical materials that have extensive utilization in many areas. It is also widely applied as pharmaceutical derivatives, such as the antipsychotic drug, anti-infective drug, and antitumor drug. However, hydrazine is a highly toxic reagent which can bring severe damage to human and animals. Herein, we have reported a new benzothiazole substituted naphthalene based probe for hydrazine with huge fluorescence change ratio. The fluorescence change ratio at F390/F509 is up to 200-fold in the presence of hydrazine in 60 min. The probe exhibited negligible cytotoxicity toward HeLa cells and was applied for hydrazine detection successfully in HeLa cells.

Automated syringe-pump-based flow-batch analysis for spectrophotometric determination of trace hexavalent chromium in water samples
摘要: In this work, we established an automated spectrophotometric method for determining hexavalent chromium in water samples using an integrated syringe pump-based environmental water analyzer (iSEA) based on the classic 1,5-diphenylcarbazide chemistry. The device was computer-controlled with programs written by LabVIEW. The effects of reagent concentration, sample salinity and foreign ion interferences were investigated. Standard solutions (GBW(E)081584-1) were used to test the accuracy of the method. When equipped with a 5 cm Z-shaped flow cell, the detection limit was 0.024 μM with sample throughput of >30 h-1. In order to meet the requirements for trace Cr(VI) determination, the system was modified with a 2.5 m liquid waveguide capillary cell (internal diameter of 0.55 mm). The sample consumption was less than 2.1 mL. The detection limit was 0.54 nM with sample throughput of 20 h-1 and good linearity with R2=0.9985 (n=110, obtained over 14 days). The two methods were successfully applied to determine Cr(VI) level in different water samples including tap water, river water, industrial wastewater and 37 brands of bottled water with spiked recovery in the range of 89.5%-117%.

Synthesis and Characterization of novel Thiophene and Carbazole-based Polymers – Optical and Electrochemical Characterization
摘要: A donor/acceptor series of carbazole copolymers, composed of alternating 2,7-linked 3,6-difluoro-9-(1-octyl-nonyl)-carbazole units and bithiophene repeated units [P1], 5,7-bis(5-bromothiophen-2-yl)-2,3-bis(4-(2-ethylhexyloxy) phenyl) thieno[3,4-b]pyrazine repeated units [P2] and bithiophene mixed with thieno[3,4-b]pyrazine repeated units [P3] have been prepared following Suzuki polymerisation procedures. The route of synthesis and characterisation techniques of this novel class of materials, together with their photo-physical and electrochemical properties are presented in this study. The polymers were characterised by 1H NMR, 13C NMR and Elemental Analysis. Molecular weights were estimated using gel permeation chromatography (GPC). The thermal stability behaviour for polymers was investigated using thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC). The electronic and photo-physical properties were investigated by use of cyclic voltammetry (CV) and UV-Vis spectroscopy, respectively.

FTIR imaging of MCF-7 colonies and their vicinity in Matrigel-embedded 3D cultures
摘要: BACKGROUND: It is now recognized that tumor cells can actively alter their microenvironment and that this remodelled microenvironment can subsequently play a critical role in cancer progression and in?uence therapeutic responses. To date, the molecular heterogeneity within a 3D cancer cell colony and its in?uence on the extracellular matrix have not been studied by infrared imaging. OBJECTIVE: The objective of this study is to investigate by mid-infrared imaging 3D Matrigel-embedded colonies of pure MCF-7 human mammary adenocarcinoma cell line and the surrounding microenvironment after undergoing formalin ?xation and paraf?n embedding (FFPE). METHODS: In order to better reproduce the procedure used for preservation and storage of clinical tissue specimens for pathologic analysis, 7- and 10-day MCF-7 colonies embedded and grown in Matrigel were FFPE-treated; 4-μm-thick sections were cut, mounted on barium ?uoride window and deparaf?nized. The Fourier transform infrared (FTIR) images of 4096 spectra were collected in transmission mode using a FPA-based FTIR imaging system. They were pre-processed and analysed by principal component and K-mean cluster analyses. RESULTS: At 1654 cm?1 and 1234 cm?1, the intensity of absorption in the colonies is signi?cantly higher than in Matrigel. It can be also noted, on the one hand, that there is a spectral heterogeneity in the intracolonial distribution of the absorbances at 1654, 1644, 1640 and 1634 cm?1 (Amide I range) possibly due to changes in protein secondary structures. On the other hand, we observe that Matrigel close to MCF-7 colonies appears altered with respect to more distant Matrigel matrix. CONCLUSIONS: FTIR imaging allowed us to highlight changes in the chemical content in MCF-7 colonies and their direct vicinity in Matrigel-embedded 3D cultures.

Pest Control Lamp in Green Food Bases Based on Wind-solar Complementary Generation System
摘要: According to the energy complementary characteristics of wind energy and solar energy in time and space, in this study, the wind turbine and solar cells are used to design a pest control lamp in green food bases powered by a small power wind-solar complementary supply system and the intelligent controller is used to charge and discharge the batteries, realizing various protection functions. Application shows that the pest control lamp in green food bases system has the advantages of low manufacturing cost and high reliability, suitable for being used in farmland and woodland which are far from the grid.
