  • Direct mineralogical imaging of economic ore and rock samples with multi-modal nonlinear optical microscopy

    摘要: Multi-modal nonlinear optical (NLO) microscopy, including stimulated Raman scattering (SRS) and second harmonic generation (SHG), was used to directly image mineralogical features of economic ore and rock samples. In SRS/SHG imaging, ore samples generally require minimal preparation and may be rapidly imaged, even in their wet state. 3D structural details, at submicron resolution, are revealed tens of microns deep within samples. Standard mineral imaging based on scanning electron microscopy (SEM), with elemental analysis via energy dispersive X-Ray spectroscopy, was used to independently validate the mineral composition of the samples. Spatially-resolved SRS from dominant Raman-resonant bands precisely maps the locations of specific minerals contained within the samples. SHG imaging reveals locally non-centrosymmetric structures, such as quartz grains. Competing absorption and nonlinear scattering processes, however, can reduce contrast in SRS imaging. Importantly, the correlation between standard electron microscopy and multi-modal NLO optical microscopy shows that the latter offers rapid image contrast based on the mineral content of the sample.

  • Correlated Materials Characterization via Multimodal Chemical and Functional Imaging

    摘要: Multimodal chemical imaging simultaneously offers high resolution chemical and physical information with nanoscale, and in select cases atomic resolution. By coupling modalities that collect physical and chemical information, we can address scientific problems in biological systems, battery and fuel cell research, catalysis, pharmaceuticals, photovoltaics, medicine and many others. The combined systems enable local correlation of material properties with chemical makeup, making fundamental questions in how chemistry and structure drive functionality approachable. In this review we present recent progress and offer a perspective for chemical imaging used to characterize a variety of samples by a number of platforms. Specifically, we present cases in infrared and Raman spectroscopies combined with scanning probe microscopy; optical microscopy and mass spectrometry; nonlinear optical microscopy; and finally, ion, electron and probe microscopies with mass spectrometry. We also discuss the challenges associated with the use of data originated by the combinatorial hardware, analysis, and machine learning as well as processing tools necessary for interpretation of multidimensional data acquired from multimodal studies.

  • Nonlinear optical microscopy for label-freely detecting gastric intestinal metaplasia

    摘要: Intestinal metaplasia has been widely considered as a precursor of gastric cancer. As a result, early detection and accurate diagnosis will have important clinical significance. Therefore, nonlinear optical microscopy using two-photon excited fluorescence combined with second harmonic generation was used for investigating gastric mucosal intestinal metaplasia in this work. The imaging results showed that this microscope has the ability to directly identify this lesion from normal tissues in the absence of labels, and to further differentiate the subtyping of intestinal metaplasia. Furthermore, nuclear areas from normal gastric mucosa and intestinal metaplasia subtypes were measured, respectively, to show the cell changes while gastric mucosa developed from normal to intestinal metaplasia. This study may provide an approach for further researching these diseases in the future, and highlights the potential of nonlinear optical microscopy as a diagnostic tool for label-free identification of gastric intestinal metaplasia.
