  • Flow cytometry for fast screening and automated risk assessment in systemic light-chain amyloidosis

    摘要: Early diagnosis and risk stratification are key to improve outcomes in light-chain (AL) amyloidosis. Here we used multidimensional-flow-cytometry (MFC) to characterize bone marrow (BM) plasma cells (PCs) from a series of 166 patients including newly-diagnosed AL amyloidosis (N = 94), MGUS (N = 20) and multiple myeloma (MM, N = 52) vs. healthy adults (N = 30). MFC detected clonality in virtually all AL amyloidosis (99%) patients. Furthermore, we developed an automated risk-stratification system based on BMPCs features, with independent prognostic impact on progression-free and overall survival of AL amyloidosis patients (hazard ratio: ≥ 2.9;P ≤ .03). Simultaneous assessment of the clonal PCs immunophenotypic protein expression profile and the BM cellular composition, mapped AL amyloidosis in the crossroad between MGUS and MM; however, lack of homogenously-positive CD56 expression, reduction of B-cell precursors and a predominantly-clonal PC compartment in the absence of an MM-like tumor PC expansion, emerged as hallmarks of AL amyloidosis (ROC-AUC = 0.74;P < .001), and might potentially be used as biomarkers for the identification of MGUS and MM patients, who are candidates for monitoring pre-symptomatic organ damage related to AL amyloidosis. Altogether, this study addressed the need for consensus on how to use flow cytometry in AL amyloidosis, and proposes a standardized MFC-based automated risk classification ready for implementation in clinical practice.

  • Sequential Growth of High Quality Sub-10 nm Core-Shell Nanocrystals: Understanding the Nucleation and Growth Process Using Dynamic Light Scattering

    摘要: Monodisperse sub-10 nm core-shell nanocrystals have been extensively studied owing to their important applications in catalysis, bioimaging, nanomedicine and so on. In this work, an amorphous shell component-crystallization strategy has been proposed to prepare high quality sub-10 nm NaYF4:Yb/Er@NaGdF4 core-shell nanocrystals successfully via a sequential growth process. Dynamic light scattering technique has been used to investigate the secondary nucleation and growth process of forming the core-shell nanocrystals. The size and morphologies evolution of the core-shell nanocrystals reveal that the secondary nucleation of the shell component is unavoidable after hot-injecting of the shell precursor at high temperature, which followed by dissolution and re-crystallization process (an Ostwald Ripening process) to partially produce the core-shell nanocrystals. The present study demonstrates that the size of seed nanocrystals and the injection temperature of the shell component precursor play a vital role in formation of core-shell nanostructures completely. This work will provide an alternative strategy for precisely controllable fabrication of sub-10nm core-shell nanostructures for various applications.

  • Viscoelastic change of block copolymer ion gels in a photo-switchable azobenzene ionic liquid triggered by light

    摘要: A photo-switchable ionic liquid solvent bearing an azobenzene moiety induced a viscoelastic change of block copolymer ion gels by light. ABA triblock copolymers having poly(phenethyl methacrylate) and poly(benzyl methacrylate) as A blocks exhibited opposite photo-induced rheological responses although they had only a tiny structural difference in the alkyl chain length.

  • Visible light thiocyanation of <i>N</i> -bearing aromatic and heteroaromatic compounds using Ag/TiO <sub/>2</sub> nanotube photocatalyst

    摘要: In this study, Ag/TiO2 nanotubes (Ag/TNT) were synthesized via simple hydrothermal process, and this photocatalyst was successfully exploited in thiocyanation reactions at room temperature under visible light irradiation. Four classes of important heterocyclic compounds including indole, aniline, pyrrole and 2-amino thiazole derivatives via Ag/TNT treated with ammonium thiocyanate formed the corresponding thiocyano compounds in moderate to excellent yields. Plasmonic property and catalytic performance of Ag nanoparticles (which mainly deposited on the surface of TNT) in absorbing species containing sulfur atoms were key components for the reactions to proceed. This potent, simple and versatile protocol reported for the first time afforded thiocyanation reactions of aniline, pyrrole and 2-amino thiazole derivatives under visible light. The reactions proceeded through a radical pathway by applying air molecular oxygen as a low-cost and environmentally friendly terminal oxidant.

  • Focal overlap gating in velocity map imaging to achieve high signal-to-noise ratio in photo-ion pump-probe experiments

    摘要: We demonstrate a technique in velocity map imaging (VMI) that allows spatial gating of the laser focal overlap region in time resolved pump-probe experiments. This significantly enhances signal-to-noise ratio by eliminating background signal arising outside the region of spatial overlap of pump and probe beams. This enhancement is achieved by tilting the laser beams with respect to the surface of the VMI electrodes which creates a gradient in flight time for particles born at different points along the beam. By suitably pulsing our microchannel plate detector, we can select particles born only where the laser beams overlap. This spatial gating in velocity map imaging can benefit nearly all photo-ion pump-probe VMI experiments especially when extreme-ultraviolet light or X-rays are involved which produce large background signals on their own.

  • Green synthesis of g-C <sub/>3</sub> N <sub/>4</sub> -Pt catalyst and application to photocatalytic hydrogen evolution from water splitting

    摘要: The g-C3N4-Pt photocatalyst was successfully prepared by the combination of a biosynthesis method and sol deposition, which were used for hydrogen evolution from water splitting. The layers of g-C3N4 are thinned and the Pt nanoparticles simultaneously become tightly bound to g-C3N4 by secondary calcination in the process of synthesizing the g-C3N4-Pt photocatalyst. Analysis of the morphological structure and instrumental characterization of the optical performance revealed that the Pt nanoparticles were successfully loaded and well dispersed on the surface of g-C3N4. Furthermore, the absorption wavelength range of the g-C3N4-Pt photocatalyst in visible light was widened and the absorption increased. The activity and photostability of the g-C3N4-Pt photocatalyst for hydrogen evolution under visible light irradiation (λ≥420 nm) were excellent. The rate of H2 evolution reached 582.4 mmol h?1 g?1, and the quantum efficiency (QE) reached 2.70% at 420 nm.

  • Light propagated distance and redshift of a distant star

    摘要: According to the formerly reported 4-D spherical model of the time and universe, factors affecting the redshift are discussed. In addition to the factor from the space expansion, two other factors derived from the light speed variation are proposed. One is the energy density factor of the wave medium, which was formerly reported to determine the light speed. The second is a newly proposed factor caused by the electromagnetic interaction of light with substances in the 3-D space. Subsequently the direct correlation of the light propagated distance with the redshift is given. Superimposed graphs of it on the real observed data from the Supernova Cosmology Project exhibited an excellent fit for a case that the current radius (equal to our observed time) of the universe is between 0.7 and 0.8 of its maximum. This could be an important ground for a possibility of the 4-D spherical model, which implies that the universe has been expanding at a constant speed by our observed time.

    摘要: Recording and imaging the 3D world has led to the use of light fields. Capturing, distributing and presenting light field data is challenging, and requires an evaluation platform. We define a framework for real-time processing, and present the design and implementation of a light field evaluation system. In order to serve as a testbed, the system is designed to be flexible, scalable, and able to model various end-to-end light field systems. This flexibility is achieved by encapsulating processes and devices in discrete framework systems. The modular capture system supports multiple camera types, general-purpose data processing, and streaming to network interfaces. The cloud system allows for parallel transcoding and distribution of streams. The presentation system encapsulates rendering and display specifics. The real-time ability was tested in a latency measurement; the capture and presentation systems process and stream frames within a 40 ms limit.

    摘要: The capturing of angular and spatial information of the scene using single camera is made possible by new emerging technology referred to as plenoptic camera. Both angular and spatial information, enable various post-processing applications, e.g. refocusing, synthetic aperture, super-resolution, and 3D scene reconstruction. In the past, multiple traditional cameras were used to capture the angular and spatial information of the scene. However, recently with the advancement in optical technology, plenoptic cameras have been introduced to capture the scene information. In a plenoptic camera, a lenslet array is placed between the main lens and the image sensor that allows multiplexing of the spatial and angular information onto a single image, also referred to as plenoptic image. The placement of the lenslet array relative to the main lens and the image sensor, results in two different optical designs of a plenoptic camera, also referred to as plenoptic 1.0 and plenoptic 2.0. In this work, we present a novel dataset captured with plenoptic 1.0 (Lytro Illum) and plenoptic 2.0 (Raytrix R29) cameras for the same scenes under the same conditions. The dataset provides the benchmark contents for various research and development activities for plenoptic images.

    摘要: Currently, available plenoptic imaging technology has limited resolution. That makes it challenging to use this technology in applications, where sharpness is essential, such as film industry. Previous attempts aimed at enhancing the spatial resolution of plenoptic light field (LF) images were based on block and patch matching inherited from classical image super-resolution, where multiple views were considered as separate frames. By contrast to these approaches, a novel super-resolution technique is proposed in this paper with a focus on exploiting estimated disparity information to reduce the matching area in the super-resolution process. We estimate the disparity information from the interpolated LR view point images (VPs). We denote our method as light field block matching super-resolution. We additionally combine our novel super-resolution method with directionally adaptive image interpolation from [1] to preserve sharpness of the high-resolution images. We prove a steady gain in the PSNR and SSIM quality of the super-resolved images for the resolution enhancement factor 8x8 as compared to the recent approaches and also to our previous work [2].
