  • SwissKnife-Inspired Multifunctional Fluorescence Probes for Cellular Organelle Targeting Based on Simple AIEgens

    摘要: Facile, efficient and mass production of aggregation-induced emission (AIE) luminogens (AIEgens) with excited-state intramolecular proton transfer (ESIPT) characteristics was achieved by one-step condensation reaction of 2-(hydrazonomethyl)phenol with benzaldehydes. The function of as-prepared AIEgens could be tuned easily by varying the functional group carrying on the phenyl ring of benzaldehyde just like Swiss knife handle. The suitable distance and angle of intramolecular hydrogen bond in these AIEgens endowed them with ESIPT properties, intense solid-state luminescence and large Stokes shifts (155?169 nm). These AIEgens could not only serve as biological probes showing specific targeting to lipid droplets, endoplasmic reticulum and lysosomes, respectively, but also generate reactive oxygen species upon visible light irradiation to make them promise for photodynamic therapy.

  • A Ratiometric Fluorescence Probe for Cisplatin: Investigating Intracellular Reduction of Platinum(IV) Prodrug Complexes

    摘要: Pt(IV) prodrug strategy has emerged as an excellent alternative to tackle the problems associated with conventional Pt(II) drug therapy. However, there is a lack of tools to study how this new class of Pt(IV) drugs are processed at the cellular level. Herein, we report the first ratiometric fluorescence probe designed for detecting cisplatin, applied to investigate Pt(IV) anticancer complexes in biological systems. The probe was able to distinguish between cisplatin and its Pt(IV) derivatives, allowing us to probe the intracellular reduction of Pt(IV) prodrug complexes. The correlation between the amount of active Pt(II) species available after intracellular reduction of Pt(IV) complexes to their cytotoxicity and the role glutathione plays in their reduction were investigated.

  • Rational structure‐based design of fluorescence probes for amyloid folds

    摘要: Amyloid fibrils are pathological hallmark of various human diseases - including Parkinson’s Alzheimer’s, ALS, Prion diseases. Treatment of the amyloid diseases are hindered amongst other factors also due to timely detection and therefore, early detection of the amyloid fibrils would be beneficial for the treatment against these disorders. Here, we report a small molecular fluorescent probe that selectively recognize the fibrillar form of Amyloid beta(1-42), a-Synuclein, and HET-s(218-289) protein over their monomeric conformation. The rational design of the reporters relies on the well-known cross-b-sheet repetition motif, the key structural feature of amyloids.
