A “turn-on” fluorometric assay for kanamycin detection by using silver nanoclusters and surface plasmon enhanced energy transfer
摘要: A rapid method is described for the determination of the antibiotic kanamycin. It integrates a kanamycin-binding aptamer and surface plasmon enhanced energy transfer (SPEET) between DNA-templated silver nanoclusters (AgNCs) and gold nanoparticles (AuNPs). The AgNCs and AuNPs were selected as energy donor and energy acceptor, respectively. The aptamer was designed to regulate the energy transfer between AgNCs and AuNPs. The aptamer was adsorbed on the AuNPs. Upon addition of kanamycin, the aptamer-kanamycin complex is formed, and this results in the aggregation of the AuNPs in high salt concentration, the formation of a blue coloration, and in the suppression of the SPEET process. The fluorescence of the AgNCs (with excitation/emission peaks at 560/600 nm) is quenched by the aptamer protected AuNPs in absence of kanamycin. The fluorescence on addition of kanamycin increases linearly in the 5 to 50 nM concentration range, with a lower detection limit of 1.0 nM (at S/N = 3). The assay can be performed within 30 min. It was successfully applied to the determination of kanamycin in spiked milk samples, and recoveries ranged between 90.2 and 95.4%. Conceivably, the strategy has a wide potential for screening by simply changing the aptamer.

Preparation of DNA functional phosphorescent quantum dots and application in melamine detection in milk
摘要: Bio-functionalization of quantum dots (QDs) is of important value in practical applications. With single-stranded DNA (ssDNA) rich in thymine T and thioguanine G taken as the template, a new-type nanocomposite material (ssDNA-PQDs) synthesized from low-toxicity T-ssDNA functionalized Mn–ZnS and room-temperature phosphorescent (RTP) QDs (PQDs) was prepared in this paper by optimizing synthesis conditions, and these ssDNA-PQDs could emit orange RTP signals at 590 nm. As these ssDNA-PQDs are rich in T sequences and T sequences can bond with melamine through the hydrogen-bond interaction, ssDNA-PQDs experience aggregation, thus causing phosphorescent exciton energy transfer (PEET) between ssDNA-PQDs of di?erent particle sizes and their RTP quenching. Based on this principle, an RTP detection method for melamine was established. The linear range and detection limit of the detection method are 0.005–6 mM and 0.0016 mM respectively. As this method is based on the RTP nature of ssDNA-PQDs, it can avoid disturbance from background ?uorescence and scattered light of the biological ?uid, and it is very suitable for melamine detection in the biological ?uid milk.

Short communication: Fourier-transform mid-infrared spectroscopy to predict coagulation and acidity traits of sheep bulk milk
摘要: Sheep milk is mainly transformed into cheese; thus, the dairy industry seeks more rapid and cost-effective methods of analysis to determine milk coagulation and acidity traits. This study aimed to assess the feasibility of Fourier-transform mid-infrared spectroscopy to determine milk coagulation and acidity traits of sheep bulk milk and to classify milk samples according to their renneting capacity. A total of 465 bulk milk samples collected in 140 single-breed flocks of Comisana (84 samples, 24 flocks) and Sarda (381 samples, 116 flocks) breeds located in Central Italy were analyzed for coagulation properties (rennet coagulation time, curd firming time, and curd firmness) and acidity traits (pH and titratable acidity) using standard laboratory procedures. Fourier-transform mid-infrared spectroscopy prediction models for these traits were built using partial least squares regression analysis and were externally validated by randomly dividing the full data set into a calibration set (75%) and a validation set (25%). The discriminant capacity of the rennet coagulation time prediction model was determined using partial least squares discriminant analysis. Prediction models were more accurate for acidity traits than for milk coagulation properties, and the ratio of prediction to deviation ranged from 1.01 (curd firmness) to 2.14 (pH). Moreover, the discriminant analysis led to an overall accuracy of 74 and 66% for the calibration and validation sets, respectively, with greater sensitivity for samples that coagulated between 10 and 20 min and greater specificity to detect early-coagulating (<10 min) and late-coagulating (20–30 min) samples. Results suggest that Fourier-transform mid-infrared spectroscopy has the potential to help the dairy sheep industry identify milk with better coagulation ability for cheese production and thus improve milk transformation efficiency. However, further research is needed before this information can be exploited at the industry level.

Ultrasensitive analysis of kanamycin residue in milk by SERS-based aptasensor
摘要: An ultrasensitive method for the kanamycin (KANA) detection in milk sample using surface-enhanced Raman spectroscopy-based aptasensor was employed in the current study. Double strand DNA binding bimetallic gold@ silver nanoparticles were developed as a sensing platform. Probe DNAs were first embedded on the surface of gold nanoparticles by the end-modified thiol, and after silver shell encapsulating, KANA aptamer DNAs with the Raman reporter Cy3 were then hybridized with probe DNAs by complementary base pairing. Results showed that with increase in the KANA concentration, the Raman intensity of Cy3 decreased. Besides achieving selectivity, an ultralow detection limit of 0.90 pg/mL, a broad linear relationship ranging from 10 μg/mL to 100 ng/mL in aqueous reagent and satisfactory recoveries of 90.4–112% in liquid whole milk were obtained. The result of actual sample proved that this aptasensor was promising in trace determination of KANA residue.

Amino-Functionalized Al-MOF for Fluorescent Detection of Tetracyclines in Milk
摘要: A fluorescent method for detection of tetracyclines (TCs) in milk was developed by using the NH2-MIL-53(Al) nanosensor synthesized via a one-pot hydrothermal method. The nanosensor had a crystalline nanoplates structure with rich groups of -NH2 and -COOH. The -NH2/-COOH of NH2-MIL-53(Al) reacted with the -CO-/-OH of TCs to form a complex. The electron of -NH2/-COOH from the NH2-BDC ligand transferred to the -CO-/-OH of TCs. -NH2 of the NH2-MIL-53(Al) interacted with the -CO-/-OH of TCs by hydrogen bonding. Quenching efficiency of inner filter effect (IFE) was calculated to contribute 57-89%. The synergistic effect of photoinduced electron transfer (PET) and IFE account for fluorescence quenching. TCs were quantitatively detected in milk samples with recoveries of 85.15 ~ 112.13%, the results were in great accordance with high performance liquid chromatography (HPLC) (P > 0.05), confirming the NH2-MIL-53(Al) nanosensor has potential applicability for the detection of TCs in food matrix.

Ultra-sensitive method based on time-resolved fluorescence immunoassay for detection of sulfamethazine in raw milk
摘要: A novel lateral flow assay (LFA) was developed by introducing Eu (III)-doped polystyrene nanoparticles (EuNPs) for rapid and ultra-sensitive detection of sulfamethazine (SM2) in raw milk. The limit of detection and linear range of the proposed method were 0.0045 and 0.05–10 ng/mL, respectively. The recovery of LFA for the detection of SM2 in raw milk was 96.1–108.2%. The proposed LFA provides a rapid and convenient strategy for fast and ultra-sensitive screening of SM2 in raw milk. EuNP-LFA may be a remarkable method for the detection of other targets at low concentrations to ensure food safety.

A new formaldehyde optical sensor: detecting milk adulteration
摘要: A sensor consisting of an optic fibre with the exposed tip coated with a the polyoxometalate salt [(C4H9)4N]4H[PMo10V2O40], specially designed to be insoluble in water, which UV-Vis spectrum changed in contact with formaldehyde, is presented. The sensor limit of detection for formaldehyde was 0.2 mg L-1, and the limit of quantification was 0.6 mg L-1, which were close to the conventional spectrophotometric method values of 0.2 mg L-1 and 0.5 mg L-1, respectively, and lower than the tolerable limit for ingested food. The sensor was tested for formaldehyde quantification in milk, as its deliberate addition is a matter of concern. The results obtained analysing formaldehyde in milk samples by the optical sensor and by the conventional method were not statistically different (α=0.05).

Interaction effect of LED color temperatures and light-protective additive packaging on photo-oxidation in milk displayed in retail dairy case
摘要: Effect of varied LED color temperatures on photo-oxidation in 2 % fat milk and protection efficiency of packaging with and without light-protective additives (LPA) under different color temperatures was firstly evaluated. Riboflavin (Rb) is one of the critical photo-sensitizers in 2 % milk when exposed to LED light. Higher color temperature with higher relative intensity of Rb absorbance region resulted in lower Rb and vitamin A retention, lower dissolved oxygen content, and higher TBARS value in milk packaged with non-LPA packaging. Yellow pigmented packaging that completely blocked the Rb absorbance region of all three LED color temperatures successfully reduced the rate of degradation of milk nutrients and flavor. TiO2-added packaging partially block the destructive light wavelength; higher level of TiO2 provided a longer protection on milk freshness. Combination of appropriate LED color temperature and LPA-packaging provided a cost-effective solution for minimizing photo-oxidation in retail dairy case.

Multi-dimensional fluorescence spectroscopy coupled with chemometrics in rapid antibiotic detection and discrimination
摘要: Antibiotic residues in animal-derived foods pose risk to human health including chemical poisoning and antimicrobial resistance, and therefore, constant monitoring throughout the food supply chain is important. In the present study multi-dimensional fluorescence spectroscopy (3D and 2D) coupled with chemometric tools were tested for potential application to detect, discriminate and quantify penicillin G (PG), sulfadiazine (SF) and tetracycline (TC) in milk by direct measurement. Qualitative and quantitative calibration models were developed for prediction of antibiotic residues. Results demonstrated optimal discrimination of milk samples on the basis of antibiotic type and concentration with close to 100% of accuracy. Negative correlation between antibiotic concentration and fluorescence peak height was displayed (r ≥ 0.963 and p ≤ 0.002). A great potential for quantitative antibiotic determination was established with R2 > 0.9 and low standard errors of estimation indicating acceptable precision for the developed technique. Fluorescence spectroscopy demonstrated high specificity and sensitivity with detection limit below the maximum residue limit of PG, SF and TC in milk. Therefore, fluorescence spectroscopy can be used as an alternative method for rapid screening of antibiotic residues in milk at collection centers and processing plants to ensure product quality and safety.

Fabrication of Milk Fat Sensor based on Plastic Optical Fiber
摘要: The research about milk fat content based on plastic optical fiber sensor using gamma and spiral spring without cladding configurations has been reported. LED light transmitted through the plastic optical fiber to be received by the phototransistor. Changes in milk fat content affect the light intensity received by the phototransistor. Sensor output voltage amplified by a differential amplifier and then the analog signal converted into the digital signal by the Arduino Uno microcontroller furthermore, the output voltage read on a computer. The milk fat content sample with a percentage of 0.773 % to 4.473 % affect the propagation of light in the core of plastic optical fiber results in power losses that occur so that the light intensity decreases and the output voltage measured becomes smaller. The results showed that the higher the milk fat content, the sensor output voltage decreases. The best measurements obtained on the spiral spring configuration with 4 windings produce value range 0.044 V, sensitivity 11.887 mV/%, and a resolution of 0.084 %. This method is effective as well as efficiently used as milk fat sensor because it has good sensitivity and resolution.
