  • In situ imaging of live-cell extracellular pH during cell apoptosis with SERS

    摘要: Extracellular pH (pHe) is an important regulating factor that determines many cellular processes, including proliferation, differentiation, and apoptosis. In our previous work, we developed 4-MPy (4-mercaptopyridine) modified Au nanoparticles as intracellular pH sensors based on surface-enhanced Raman spectroscopy (SERS). We herein modified Au nanoparticles-assembled solid SERS substrate with 4-MPy molecules for in situ pHe sensing during apoptosis. We found a more acidic extracellular environment of cancer cells than that of normal cells from the pH imaging. We then in situ investigated the temporal and spatial evolution of pHe of cancer cells after addition of transforming growth factor-β (TGF-β). The pHe showed a fast decrease at the beginning, followed by a slow decrease until the complete loss of cellular functions and the pH values in and out of the cells became similar. This work shows that our SERS substrate combined with in situ cell culture system is well suitable for in situ pHe sensing during cell processes and will be a promising technique for understanding more pHe-related biological and pathological issues.

  • The effects of 308-nm excimer laser on the infiltration of CD4+, CD8+ T-cells and regulatory T cells in the lesional skin of patients at active and stable stages of nonsegmental vitiligo

    摘要: Background: To explore the associations between this treatment and CD4+, CD8+ T-cells and regulatory T cells (Treg). Methods: We collected the skin biopsies from 295 patients with nonsegmental vitiligo treated with or without 308-nm excimer laser. We revealed that patients at stable stage showed a better response to 308-nm excimer laser than those at active stage. Results: In comparison with untreated patients, CD4+ and CD8+ T cells were both reduced while Foxp3+ Treg cells, TGF‐ β and IL-10 were elevated in the lesional sites of patients who showed effective reaction to the treatment. In the treated patients at active stage, the infiltration of CD4+ and CD8+ T cells were significantly declined but Foxp3+ Treg cells, TGF‐ β and IL-10 were increased compared to those in untreated patients at active stage. These changes of cell infiltration were more obvious in the treated patients at stable stage, explaining why there were more patients at stable stage response better than patients at active stage. Conclusion: 308-nm excimer laser is effective to reduce the infiltration of CD4+ and CD8+ T cells but promote the infiltration Treg cells and secretion of TGF‐ β and IL-10 in the lesion skin of vitiligo patients, especially at stable stage.

  • Effects of Pulsed 810?nm Al‐Ga‐As Diode Laser on Wound Healing Under Immunosuppression: A Molecular Insight

    摘要: Background and Objectives: Dysregulated inflammation is one of the major contributing factors for the prevalence of non‐healing chronic wound in immunosuppressed subjects. Photobiomodulation (PBM) has emerged as a potential non‐thermal, light‐based therapeutic healing intervention for the treatment of impaired wounds. Study Design/Materials and Methods: The present study delineates the underlying molecular mechanisms of PBM 810 nm laser‐induced full‐thickness cutaneous wound repair in immunosuppressed rats at continuous and pulsed wave‐mode with power‐density of 40 mW/cm2, fluence 22.6 J/cm2 for 10 minutes daily for 7 post‐wounding days. Molecular markers were assessed using biochemical, assay quantification, enzyme kinetics and immunoblots analyses pertaining to inflammation, oxidative stress, cell survival, calcium signaling, and proliferation cascades. Results: Results distinctly revealed that pulsed 810 nm (10 Hz) PBM potentially influenced the cell survival and proliferation signaling pathway by significantly upregulated phospho‐protein kinase B(phospho‐Akt), phospho‐extracellular‐signal‐regulated kinase 1 (ERK1), transient receptor potential vanilloid‐3 (TRPV3), Ca2+, calmodulin, transforming growth factor‐β1 (TGF‐β1), TGF‐βR3, and Na+/K+‐ATPase pump levels. PBM treatment resulted in reduction of exaggerated inflammatory responses evident by significantly repressed levels of interleukin‐1β (IL‐1β), IL‐6, cyclooxygenase 2 (COX‐2), and substance‐P receptor (SPR), as well as inhibited apoptotic cell death by decreasing p53, cytochrome C, and caspase 3 levels (P < 0.05), which, in turn, effectively augment the wound repair in immunosuppressed rats. PBM treatment also lowered 4‐hydroxynoneal (HNE) adduct level and NADP/NADPH ratio and upregulated the GRP78 expression, which might culminate into reduced oxidative stress and maintained the redox homeostasis. Conclusions: Taken together, these findings would be helpful in better understanding of the molecular aspects involved in pulsed 810 nm laser‐mediated dermal wound healing in immunosuppressed rats through regulation of cell survival and proliferation via Ca2+‐calmodulin, Akt, ERK, and redox signaling.

  • Serum TGF-?2 and TNF-?± During Psoriasis Therapy with Narrowband Ultraviolet B

    摘要: Although it is now an accepted concept that narrow-band UVB therapy is an efficacious therapy for psoriasis, the relationship between the response rate and the potential effects on serum cytokines is less well-established. The purpose of this study was to investigate the correlation between the response rate and the changes of serum TGF-β and TNF-α necessary for understanding the underlying mechanisms of narrow-band UVB phototherapy. NB-UVB is effective against psoriasis without any obvious side effects and can significantly decrease serum TNF-α and promote TGF-β level of psoriasis patients. Additionally, measurement of TGF-β and TNF-α in serum could be considered as biomarkers of psoriasis activity during NB-UVB therapy.
