  • Beyond DPPH: Use of Fluorescence-Enabled Inhibited Autoxidation to Predict Oxidative Cell Death Rescue

    摘要: Shah et al. develop a fluorescence-enabled inhibited autoxidation (FENIX) approach that permits accurate quantitation of radical-trapping antioxidant activity in phospholipid bilayers. The methodology is far superior to existing assays (e.g., DPPH) and enables reliable prediction of the anti-ferroptotic potency of redox-active compounds. The approach is easily amenable to high-throughput screening.

  • Accelerating the Screening of Perovskite Compositions for Photovoltaic Applications through High‐Throughput Inkjet Printing

    摘要: The exploration and optimization of numerous mixed perovskite compositions are causing a strong demand for high-throughput synthesis. Nevertheless high-throughput fabrication of perovskite films with representative film properties, which can efficiently screen the perovskite compositions for photovoltaic applications, has rarely been explored. A high-throughput inkjet printing approach that can automatically fabricate perovskite films with various compositions with high reproducibility and high speed is developed. The automatic sequential printing of four precursors forms 25 mixed films in a fast and reproducible manner. The obtained bandgaps, photoluminescence (PL) peak positions, and PL lifetimes allow for the efficient screening of perovskite compositions for photovoltaic applications. To exemplify this concept, among 25 tested films, two compositions CH3NH3PbBr0.75I2.25 (MA) and (HC(NH2)2)0.75(CH3NH3)0.25PbBr0.75I2.25 (FA0.75MA0.25) with a long (237 ns) and short (49.0 ns) PL lifetime, respectively, are screened out for device investigations. As expected, the MA-based device exhibits a much higher efficiency (19.0%) than that (15.3%) of the FA0.75MA0.25 counterpart. This efficiency improvement is mainly ascribed to a smaller dark saturate current density, a lower level of energetic disorder, more efficient charge transfer and decreased charge recombination losses, which are consistent with the much longer PL lifetime in the database.

  • Optimal SVC placement for Maximizing Photovoltaic Hosting Capacity in Distribution Network

    摘要: High throughput phenotyping holds promise of accelerating crop breeding programs and has gained great attention in the past several years. Although many platforms for data collection, including ground and aerial vehicles, have been developed to adapt to different crops and field setups, fully autonomous ground robots have not been readily available and widely used so far. In this study, we developed a ground robot that is capable of autonomously navigating in the field. We designed a sensor mounting frame that can be modified to mount different sensors for different data collection tasks. The robot was tested in a cotton breeding field and color images of the plants were collected. We developed an algorithm to count the cotton bolls using the color images. The preliminary results demonstrated that the system was capable of collecting data autonomously without human intervention and the collected data were useful to extract phenotypic traits.

  • Fluorescent labelling in living dental pulp stem cells by graphene oxide quantum dots

    摘要: Bighead carp (Aristichthys nobilis) and silver carp (Hypophthalmichthys molitrix) are closely related species in the subfamily Xenocypridinae within Cyprinidae, and they are also two of the four most important pond-cultured fish species in China. The ability to resist some diseases often differs significantly in silver carp and bighead carp during fishery production. However, the evolutionary divergence of the immune defense functions in these two species is still not understood at the molecular level. The data presented in this article are related to the research article entitled “Comparative analysis of spleen transcriptome detects differences in evolutionary adaptation of immune defense functions in bighead carp and silver carp” (Li et al., 2018). Please refer to this data article for interpretation of the data. Data provided in this submission comprise the Ka/Ks ratios of orthologs as well as adaptive evolution genes, expression levels of orthologs, and TPM value of genes expressed only in spleen of bighead carp or silver carp. These data provide a better understanding of the differences in evolutionary adaptation of immune defense functions in bighead carp and silver carp.

  • High-throughput screening of laser additive manufactured metallic glass via ultrasonic wave

    摘要: Laser additive manufacturing (LAM) technology provides an opportunity to fabricate bulk metallic glasses (BMGs) without any dimensional constraint and achieve the large-scale applications of BMGs. However, flaws, such as cracks, gas porosity, and crystalline phases, are always formed accompanied by the process of LAM, which seriously worsens the mechanical and physical properties of the resulting BMGs. Here, we present a novel method that involves ultrasonic wave technique to high-throughput screen the optimum process parameters for the LAM of BMG. A parameter library, constituted by a series of rectangular BMG samples, is rapidly fabricated by the LAM method under continuously changed combinations of laser power and travel speed. the ultrasonic attenuation factor, which is sensitive to the flaws, is used as the monitor to screen the parameters of the BMGs fabricated by the LAM. Using this approach, the laser power of 1300 W and travel speed of 600 mm/min are estimated as the optimum parameter combination for the LAM of a Zr51ti5ni10cu25Al9 (Zr51) BMG with the slightest flaws. The amorphous-phase dominated microstructure and the sufficiently high tensile strength of the subsequent fabricated large-sized Zr51 BMG sample verify this optimum parameter combination.

  • Exploring materials band structure space with unsupervised machine learning

    摘要: An unsupervised machine learning algorithm is applied for the first time to explore the space of materials electronic band structures. T-student stochastic neighbor embedding (t-SNE), a state of the art algorithm for visualization of high dimensional data, is applied on feature spaces constructed by extracting electronic fingerprints straight from Brillouin zone of the materials. Different spaces are designed and mapped to lower dimensions allowing to analyze and explore this previously uncharted band structure space for thousands of materials at once. In all cases analyzed machine learning was able to learn and cluster the materials depending on the features involved. t-SNE promises to be a extremely useful tool for exploring the materials space.

  • Automated high-throughput light-sheet fluorescence microscopy of larval zebrafish

    摘要: Light sheet fluorescence microscopy enables fast, minimally phototoxic, three-dimensional imaging of live specimens, but is currently limited by low throughput and tedious sample preparation. Here, we describe an automated high-throughput light sheet fluorescence microscope in which specimens are positioned by and imaged within a fluidic system integrated with the sheet excitation and detection optics. We demonstrate the ability of the instrument to rapidly examine live specimens with minimal manual intervention by imaging fluorescent neutrophils over a nearly 0.3 mm3 volume in dozens of larval zebrafish. In addition to revealing considerable inter-individual variability in neutrophil number, known previously from labor-intensive methods, three-dimensional imaging allows assessment of the correlation between the bulk measure of total cellular fluorescence and the spatially resolved measure of actual neutrophil number per animal. We suggest that our simple experimental design should considerably expand the scope and impact of light sheet imaging in the life sciences.

  • A simple turn-on fluorescent chemosensor for CO2 based on aggregation-induced emission: Application as a CO2 absorbent screening method

    摘要: An a-cyanostilbene-based fluorescent chemosensor, which includes a primary amine, was developed for the quantitative analysis of CO2 in solution. (Z)-3-(4-(3-Aminopropoxy)phenyl)-2-(4-nitrophenyl)acrylonitrile (1) reacts with CO2 and enables a turn-on type CO2 chemosensor with increased fluorescence due to aggregation-induced emission. More importantly, 1 required no additives such as an amine media for detecting CO2, and the limit of detection was estimated to be ~26 ppm. This system was successfully applied to determine the relative efficiency of CO2 absorbents dependent on the fluorescence change in 1. The use of this chemosensor as a CO2 absorbent screening method suggests that 1 may be used to develop a high-throughput screening method for the evaluation of high-performance CO2 absorbents.

  • A fluorescent 3D cell culture assay for high throughput screening of cancer drugs down-regulating survivin

    摘要: Survivin, a member of inhibitor of apoptosis family, is currently undergoing intensive investigations as a promising cancer marker due to its overexpression in multiple tumor tissues and close relationship with chemotherapy resistance. In this study, a novel 3D survivin promoter assay was developed, using enhanced green fluorescent protein (EGFP) as the reporter to assess survivin promoter activity for cancer drug screening. Breast cancer MCF-7 cells were engineered to express EGFP controlled by a human survivin promoter and a CMV promoter, respectively. These cells were cultured in three-dimensional (3D) polymer-based scaffolds on a 40-microbioreactor platform (40-MBR) with real-time monitoring of EGFP signals. The EGFP production driven by the survivin promoter was strongly correlated with survivin transcriptional level in MCF-7 cells treated with YM155, a small-molecule survivin promoter suppressant. Moreover, the potential inhibition effects of doxorubicin and cisplatin on survivin and their cytotoxicity were also evaluated in this system. This study demonstrated the potential application of the novel 3D survivin promoter-EGFP reporter assay for high-throughput screening of chemicals down-regulating survivin as a molecular target for cancer therapy.

  • Re-evaluating TiCl4 and UV assays for detection of vicine and convicine in high-throughput screening of immature and mature seeds of faba bean

    摘要: Genomic resources have just started to focus on the faba bean; the genes for synthesis of vicine and convicine (V-C) have not been determined, and recently developed genetic markers for these anti-nutritionals have not been used to examine these traits in very large scale in faba bean. Simple, rapid and cost-effective technologies are crucial in crop breeding programs, especially in the developing world, and in some cases, traditional methodologies are used in combination with genetic markers to assess agronomic traits and the value of gene markers. Here, two methodologies (TiCl4 assay and 274 nm absorption) are re-evaluated for their application in detection of V-C in faba beans. In comparison with TiCl4 assay, the method of 274 nm UV absorption without an HPLC analysis offers more reliable analysis for detection of V-C in immature and mature seeds of faba bean. Its application in high throughput screening by 60 min agitation of immature seeds or mature seed flour in 2% trichloroacetic acid (TCA) allows quick screening of low V-C faba beans. The level of V-C was maximum when seed moisture was 80% and V-C level was measured as 0.92% in CDC Fatima flour. Though V-C from 2% TCA extract of mature and immature seeds of CDC Fatima was detected by 274 wavelength in the TiCl4 assay reaction, a Ti-aglycone complex was not clearly detectible at 480 nm as previously suggested.
