  • A fluorometric method for determination of the activity of T4 polynucleotide kinase by using a DNA-templated silver nanocluster probe

    摘要: The authors describe a turn-off fluorometric method for the determination of the activity of the T4 polynucleotide kinase (T4 PNK). It is based on the use of DNA-templated silver nanoclusters (AgNCs). DNA probes with terminal 5′ hydroxy groups are used as substrates for DNA phosphatases. If subsequently treated with T4 PNK and Lambda exonuclease (λ exo), the AgNC DNA probes with a modified C-rich sequence and the G-rich sequence is separated. Upon their separation, the strong fluorescence (with excitation/emission maxima at 580/650 nm) that is caused by the proximity of the G-rich region and the C-rich region in the AgNCs decreases sharply. This enabled the fluorometric kinetic determination of the activity of T4 PNK. The assay is characterized by a wide linear range (from 0.01 to 12.5 U·mL?1), a low detection limit (0.01 U·mL?1) and short assay time (typically 60 min). This makes it a promising tool for use in studying processes related to DNA phosphorylation, in drug discovery and in diagnostics.

  • Unusual Electronic States and Superconducting Proximity Effect of Bi Films Modulated by NbSe2 Substrate

    摘要: Heterostructures of two-dimensional layered materials can be functionalized with exotic phenomena that are unpresented with each constituting component. The interface effect plays a key role in determining the electronic properties of the heterostructure, whose characterization requires a correlation with the morphology with atomic-scale precision. Here, we report an investigation on the electronic properties of few-layer Bi(110) films mediated by NbSe2 substrate. By utilizing scanning tunneling microscopy and spectroscopy, we show a significant variation of the density of states at different Bi film thicknesses, resulting in an unusual superconducting proximity effect that deviates from the conventional monotonous decay behavior. Moreover, the electronic states of the Bi films are also prominently modulated by the Moiré pattern spatially. With first-principles calculations, we illuminate these findings as the results of covalent-like quasi-bonds formed at the Bi/NbSe2 interface, which profoundly alter the charge distributions in the Bi films. Our study indicates a viable way of modulating the electronic properties of ultrathin films by quasi-covalent interfacial couplings beyond conventional van der Waals interactions.

  • Short range proximity effect induced by exchange interaction in tunnel-coupled CdTe and (Cd,Mn)Te quantum wells

    摘要: The coherent spin dynamics of electrons in tunnel-coupled CdTe and (Cd,Mn)Te quantum wells (QWs) is studied by time-resolved pump-probe Kerr rotation. The coupled QWs have different thicknesses; the narrow one is doped by Mn2+ magnetic ions. A short range proximity effect between them is observed: the Zeeman splitting of electrons in the wide QW is given in addition to the intrinsic electron g factor by the exchange interaction with the Mn2+ ions mediated by electron tunneling into the narrow QW. The exchange interaction strength scales with the Cd0.88Mg0.12Te barrier thickness separating the QWs. The Kerr rotation signal measured on the wide QW shows two close frequencies of electron spin Larmor precession in a transverse magnetic field. These components have very different spin dephasing times, 50 ps and 1 ns. The two frequencies originate from electrons in the wide QW being either part of an exciton or being resident. The proximity effect of the exciton electron is smaller due to the binding by Coulomb interaction, which decreases the tunneling to the narrow well. The experimental data are in good agreement with model calculations.

    摘要: The weak intrinsic spin-orbit coupling in graphene can be greatly enhanced by proximity coupling. Here, we report on the proximity-induced spin-orbit coupling in graphene transferred by hexagonal boron nitride (hBN) onto the topological insulator Bi1.5Sb0.5Te1.7Se1.3 (BSTS) which was grown on a hBN substrate by vapor solid synthesis. Phase coherent transport measurements, revealing weak localization, allow us to extract the carrier density-dependent phase coherence length lφ. While lφ increases with increasing carrier density in the hBN/graphene/hBN reference sample, it decreases in graphene/BSTS due to the proximity coupling of BSTS to graphene. The latter behavior results from D’yakonov-Perel’-type spin scattering in graphene with a large proximity-induced spin-orbit coupling strength of at least 2.5 meV.

  • Electronic properties of Pb islands on graphene: Consequences of a weak interface coupling from a combined STM and <i>ab initio</i> study

    摘要: By means of scanning tunneling microscopy and spectroscopy, we investigate the electronic properties of lead islands (width 5–100 nm, thickness 5–25 monolayers) deposited by molecular beam epitaxy on twisted graphene layers grown on SiC(000-1). We ?nd that elastic scattering processes govern the local density of states probed at the surface of the Pb islands, inducing (i) the well-known quantum well states due to electron con?nement in the direction perpendicular to the surface and (ii) spatial in-plane periodic modulations related to quasiparticle interferences off the island edges. Through a quantitative analysis of these effects, compared with ab initio calculations for a two-dimensional Pb slab, we conclude that the lead islands grown on the surface of graphene can be considered as freestanding from the point of view of their electronic structure, leaving the surrounding graphene layer unperturbed. Accordingly, low bias tunneling spectra show evidence of a sizable interface resistance. Nevertheless, we suggest that the transparency of the interface, which can be estimated from its resistance, is good enough to induce superconductivity within the underlying graphene layer by proximity effect with the Pb islands.

  • Non-equilibrium photoexcited carrier effects in a graphene-based Josephson junction

    摘要: We studied the superconducting proximity effect under photoexcitation by illuminating a superconductor/monolayer graphene/superconductor (SGS) Josephson junction with monochromatic light at a wavelength of 1.31 lm. Although the critical current Ic can be controlled by the irradiation power P, its variation cannot be explained by modi?cation of the carrier density, which has been reported for semiconductor-based Josephson junctions. The estimated electron temperature of gra- phene is proportional to Pd, where d ’ 1=3. This relation clearly indicates that photogenerated non- equilibrium carrier dynamics are responsible for the variation of Ic with P. We suggest that the SGS junction can directly mediate interactions between the optical ?eld and the superconducting state.
