研究目的
研究细胞因子激活和氧化应激对人胚胎干细胞来源的视网膜色素上皮细胞功能的影响，重点关注基质金属蛋白酶（MMP）表达及屏障特性，以建立年龄相关性黄斑变性等视网膜疾病的模型。

成熟的人胚胎干细胞来源视网膜色素上皮（hESC-RPE）细胞在基质金属蛋白酶表达和屏障功能方面对细胞因子及氧化应激的反应与天然RPE细胞相似，这使其适合用于体外模拟年龄相关性黄斑变性、糖尿病视网膜病变和增生性玻璃体视网膜病变等视网膜疾病。
研究不足
该研究采用体外细胞培养，可能无法完全模拟体内环境；观察到不同细胞系间存在差异；且对部分基质金属蛋白酶（MMPs）的蛋白质分泌影响未呈现统计学显著性或与基因表达变化不一致。

研究将成熟hESC-RPE细胞暴露于炎症细胞因子（IFN-γ或TNF-α）24小时或氧化应激（H2O2）1小时，随后通过TEER、qRT-PCR、Western blot、ELISA和酶谱法分析屏障功能、基因表达及蛋白分泌情况。
