Investigating the in vivo comparison of N-11CH3 vs O-11CH3 radiolabeled microtubule targeted PET ligands for imaging microtubules in the brain.

MicroPET images of MT tracers, MPC-6827 and HD-800 (radiolabeled as O-11CH3 and N-11CH3), showed BBB penetration, retention of activity in brain with high specific binding. Biodistribution data further confirmed BBB penetration and high brain uptake for both O-11CH3 and N-11CH3 labeled MT tracers. All tracers exhibit comparable brain to muscle ratios, demonstrating that the N-11CH3 labeling of the MT targeted ligands retained the in vivo binding characteristic of the corresponding O-11CH3 labeled tracers [11C]MPC-6827 and [11C]HD-800 in mice brain.

The study was conducted in mice, and tracer quantification with arterial input functions measurement is not practical in mice. Further in vivo stability measurement along with binding parameter quantification of these tracers in nonhuman primates is needed for a head-to-head comparison.
