研究目的
To develop a sensitive and selective method for the detection of thrombin and ATP using green fluorescent carbon quantum dots functionalized with polyethyleneimine (PEI-CDs) and aptamers.
研究成果
The PEI-CDs-based aptasensor provides a sensitive and selective method for the detection of thrombin and ATP. The method is applicable in complex biological samples and offers a generally applicable approach for various aptamer-specific binding targets.
研究不足
The method has a higher limit of detection (LOD) compared to some recently reported electrochemical and FRET methods. Real sample analysis using this method should be further investigated.
1:Experimental Design and Method Selection
The PEI-CDs were synthesized via hydrothermal method using malic acid and PEI as precursors. The fluorescence properties of PEI-CDs were optimized by varying the ratio of malic acid to PEI, reaction temperature, and time.
2:Sample Selection and Data Sources
Thrombin and ATP aptamers were synthesized and purified. Serum samples were used for real sample analysis.
3:List of Experimental Equipment and Materials
FL-7000 spectrofluorometer, Lambda 35 UV-Vis spectrophotometer, JEM-2100 TEM, Zetasizer Nano Z, FT-IR spectrophotometer.
4:Experimental Procedures and Operational Workflow
PEI-CDs were synthesized by hydrothermal treatment. For thrombin and ATP detection, aptamers were mixed with PEI-CDs, and the fluorescence change was measured upon addition of thrombin or ATP.
5:Data Analysis Methods
Fluorescence intensity was measured to determine the concentration of thrombin and ATP. The specificity of the aptasensor was evaluated against non-target proteins and analogs.
