THz TDS is a powerful technique to distinguish molecule structure of materials. The study of vibration spectrum of amino acids helps to understand the correlation between molecular structure and function. The main purpose of spectral research is to obtain the characteristic spectrum of the object of study.

The main difficulty in interpreting the terahertz spectrum lies in the lack of the corresponding molecular vibration model in the terahertz band. The simulation method is not yet mature, and the experimental conditions differ from the calculation conditions (vacuum vs. air).
1:Experimental Design and Method Selection:
The study utilized terahertz time-domain spectroscopy (TDS) technique to measure the absorption spectra of amino acids. The spectra were compared with theoretical simulations obtained using density functional theory (DFT).
2:Sample Selection and Data Sources:
The amino acids (L-alanine, D-alanine, DL-alanine, L-methionine, D-methionine, DL-methionine, L-valine, D-valine, DL-valine, L-leucine, D-leucine, and DL-leucine) were purchased from Sigma-Aldrich. Samples were prepared by grinding the crystals into powder and mixing with polyethylene (PE) at different ratios before compressing into tablets.

Terahertz time-domain spectrometer based on a pulsed femtosecond laser, mortar and pestle for grinding, polyethylene (PE) for mixing, and a press for tablet formation.
4:Experimental Procedures and Operational Workflow:
The absorption spectra were measured at room temperature. The samples were tested twice in different locations. The reference signal was the pulse signal in air.
5:Data Analysis Methods:
The refractive index and absorption coefficient were derived from the electric field of the sample and reference, using specific formulas to analyze the data.
