To evaluate the efficacy of photodynamic therapy (PDT) in combination treatment with colistin against pandrug-resistant Acinetobacter baumannii (PDRAB).

PDT in combination with colistin is highly effective against PDRAB, killing all treated bacterial cells and reducing the minimal inhibitory concentration of colistin by more than 11 fold. This strategy could be used against drug-resistant bacteria in localized infections, but further studies are needed to determine its clinical efficacy.

The study was conducted in vitro, and its efficacy in clinical settings needs further investigation. The mechanisms of synergy between PDT and colistin are not fully understood.
1:Experimental Design and Method Selection:
PDT was carried out using toluidine blue O (TBO) as a photosensitizer and a light-emitting diode (LED) as a radiation source. The effect of PDT plus colistin was evaluated on colistin-resistant A. baumannii (CoRAB).

A PDRAB isolated from burn patients was used as a test strain.

Bacterial suspensions were treated with TBO and exposed to LED. The effect of PDT plus colistin was evaluated, and colony-forming units were calculated.

Killing percentage and log reduction were calculated. SEM was used for confirmation of antibacterial activity.
