研究目的
To review the recent advances of FA/FP assays involving aptamers as affinity ligands for small molecule detection, focusing on aptamer-based fluorescence anisotropy assays for the detection of small molecules.
研究成果
The combination of aptamers and FA techniques has enabled the development of diverse assays for small molecule detection, benefiting from the unique properties of aptamers. Direct and competitive assays, along with signal amplification strategies, have been successfully applied. However, the expansion of these assays to more targets requires the development of new aptamers with high specificity and affinity.
研究不足
The main limitation is the need for fluorescent tracers in competitive FA assays, which can be challenging to conjugate to small molecules without disrupting their binding affinity to aptamers. Additionally, the availability of high-quality aptamers for a wide range of small molecules is limited.
