1:Experimental Design and Method Selection:
Whole-cell voltage-clamp recordings were used to study release properties of cone photoreceptors in wild-type and mutant mouse retinae with a genetic disruption of the presynaptic cytomatrix protein Bassoon. Light and electron microscopy were employed to reveal the distribution of Ca2+ channels and synaptic vesicles.
2:Sample Selection and Data Sources:
Wild-type and mutant mouse retinae were used. Cone photoreceptors were identified by their expression of eGFP in Rac3-eGFP mice.
3:List of Experimental Equipment and Materials:
Patch-clamp recordings were carried out with an EPC-10 patch-clamp amplifier. The extracellular solution contained NaCl, NaHCO3, NaH2PO4, KCl, CaCl2, MgCl2, glucose, and HEPES. The intracellular solution contained Cs-gluconate, EGTA, TEA-Cl, HEPES, Mg-ATP, and GTP.
4:Experimental Procedures and Operational Workflow:
Whole-cell recordings were carried out at room temperature. Retinal slices were observed with a 63x water immersion objective. Currents were recorded, low-pass filtered, and digitized.
5:Data Analysis Methods:
Data were analysed with Fitmaster and OriginPro. The analysis of tonic postsynaptic current was carried out with the Mini Analysis software package.
