1:Experimental Design and Method Selection:
The study integrates the peroxidase-like catalytic and fluorescent properties of GSH-AuNCs to create a ratiometric nanosensor. GSH-AuNCs catalyze the decomposition of H2O2 into hydroxyl radicals, which oxidize terephthalic acid (TA) to fluorescent hydroxyterephthalate (TAOH). The fluorescence at 430 nm (from TAOH) and 600 nm (from GSH-AuNCs) is measured upon excitation at 315 nm for self-calibration.
2:Sample Selection and Data Sources:
H2O2 and glucose solutions were prepared from analytical grade chemicals. Human serum samples were obtained from a hospital and diluted for analysis.
3:List of Experimental Equipment and Materials:
Chemicals include H2O2, glucose, TA, NaOH, NaH2PO4·2H2O, Na2HPO4·12H2O, HAuCl4·4H2O, GOx, GSH. Equipment includes FluoroMax-4 spectrofluorophotometer, UV-1800 spectrophotometer, Tecnai G2 F20 TEM, SU8220 SEM, Zetasizer Nano ZSE DLS, PB-10 pH meter.
4:Experimental Procedures and Operational Workflow:
GSH-AuNCs were synthesized by reacting HAuCl4 and GSH at 70°C for 24 hours. For H2O2 assay, H2O2 solutions, TA solution, and diluted GSH-AuNCs were mixed in phosphate buffer, incubated at 37°C for 30 min, and fluorescence spectra recorded. For glucose assay, glucose solutions, GOx, TA, and GSH-AuNCs were mixed similarly.
5:Data Analysis Methods:
Fluorescence intensity ratios (F430/F600) were calculated and correlated with concentrations using linear regression. Detection limits were based on 3 times standard deviation of blank measurements.
