  • Coumarin-based Fluorescent Probes for Super-resolution and Dynamic Tracking of Lipid Droplets

    摘要: Visualizing and dynamic tracking lipid droplets (LDs) are of great importance to biological research. Herein, two-photon absorption fluorescent small bio-probes based on lipophilic coumarin were developed, which exhibited high selectivity towards LDs in Hela cells. Owing to the good biocompatibility and excellent photo-stability, the probes were applied to realize specific super-resolution visualization of the intracellular LDs in Hela cells, offering us the quantitative results of the amount and diameters of LDs as well. Furthermore, the bio-probes were capable of monitoring the movements of the LDs in real-time. We believe that the bio-probes would provide new avenues to designing bio-imaging and biological diagnosis.

  • Rotation and translation dynamics of coumarin 153 in choline chloride-based deep eutectic solvents

    摘要: The equilibrium and dynamic solvation responses of coumarin 153 (C153) in a range of deep eutectic solvents (DESs) based on choline chloride with either urea (molar ratio 1:2, ChCl:U), glycerol (1:2, ChCl:G), ethylene glycol (1:2, ChCl:E), or malonic acid (1:1, ChCl:Mal) were investigated using both steady-state and time-resolved ?uorescence emission spectroscopy at room temperature (298 K). From steady-state ?uorescence data, “red-edge effects” were observed in all the DESs studied, attributed to spatial heterogeneity of the DES matrix. Time-resolved Stokes shifts were used to quantify dynamic solvation with the solvation response function in DES found to be a biexponential function of time, which were used to obtain average solvation times ((cid:104)τs(cid:105)) which are generally faster in DES than in ionic liquids of comparable viscosity. Average solvation times showed a partial correlation with viscosity between different DESs. The choline chloride-glycerol DES showed deviation from the viscosity trend observed in the other DES for both dynamic and steady-state results. Rotational reorientation times obtained from dynamic anisotropy (r(t)) measured for these DESs showed a partial correlation with viscosity between different DESs. Determination of the DES rotational coupling with C153 showed more “slip”-like behavior than the previously reported ionic liquids and dipolar solvents.

  • Novel nonlinear optical chromophores based on coumarin: Synthesis and properties studies

    摘要: Novel nonlinear optical chromophores JC-TCF and JC-FTCF using tetramethyljulolidine-based coumarin as the electron-donor and tricyanofuran derivatives as the electron acceptors were designed, synthesized and investigated. The obtained chromophores own good solubility in common solvents, and good compatibility with host polymers. The chromophores also have good thermal stability, and the decomposition temperature exceed 200 °C. The UV–vis spectra indicated that JC-TCF and JC-FTCF possess good intramolecular charge transfer ability. Density functional theory (DFT) was used to study the energy gap and ?rst-order hyperpolarizability(β). After contact poling, the guest-host polymer ?lms of polymethyl methacrylate containing 20% wt JC-TCF and 20% wt JC-FTCF exhibited the highest electro-optic coe?cient (r33) up to 40 p.m./V and 76 p.m./V respectively, and the results indicated the chromophores’ microscopic β value could be e?ectively translated to macroscopic EO activity, and the tetramethyljulolidine-based coumarin could be an useful candidate in the development of NLO materials.

  • A novel OFF-ON-OFF fluorescence probe based on coumarin for Al3+ and F? detection and bioimaging in living cells

    摘要: A novel fluorescence probe L2 based on coumarin has been designed and synthesized. The probe L2 can be used for relay recognition of metal ions Al3+ and anion F? in the aqueous HEPES buffer (0.05 M, pH=7.4), and build a OFF-ON-OFF detection system. The probe showed high selectivity and sensitivity to target ions in the process of relay recognition, and the corresponding detection limit could be as low as 0.014 μM (Al3+) and 0.03 μM (F?). Besides, the geometry optimizations of probe L2 and [L2+Al3+] complex were carried out using the Gaussian 16 program based on DFT, and the identification mechanism of the probe was also discussed by the mass spectrometry and theoretical calculations. Moreover, the probe have also been successfully applied to detection of target ions in living cells.

  • Insight into the Dynamics of Different Fluorophores in the Interior of Aerosol OT Lamellar Structures in Presence of Sugars: From Picosecond-to-Femtosecond Study

    摘要: It is well reported that sugar molecules provide different types of stabilization to the biomembranes both in vitro as well as in vivo. In the present article, our focus is to investigate the interactions of two sugar molecules (sucrose and sucralose) with the lamellar structures of aerosol OT (AOT). We have attempted the structural characterization of the lamellae in presence and absence of the sugars with the aid of dynamic light scattering (DLS), small-angle X-ray scattering (SAXS), Fourier transformed infrared spectroscopy (FTIR), polarized optical microscopy (POM) and cryogenic-transmission electron microscopic (Cryo-TEM) techniques. In this study, we have chosen three Coumarin dyes, which show a wide variation of hydrophobicity and performed a combination of steady-state and time-resolved fluorescence measurements to unveil the inner detail of the location of the sugars and their specific interactions with the lamellar structures. Our study reveals that sucrose molecules are present in the interfacial region with a major population whereas the most probable location of sucralose is the interior of the AOT bilayer. Therefore, sucralose molecules probably penetrate the bilayer by decreasing the efficient packing of AOT. The important essence of this study is the location and the interactions of sucralose with the lamellae which may provide a future direction to the transportation of the drug molecules in the biomembrane.
