  • On-demand CO release for amplification of chemotherapy by MOF functionalized magnetic carbon nanoparticles with NIR irradiation

    摘要: Carbon monoxide (CO) gas therapy combined with chemotherapy and photothermal therapy (PTT) is a promising treatment mode for malignant tumor. Herein, we firstly reported doxorubicin (DOX) loaded Mn carbonyl modified Fe (III)-based nanoMOFs (MIL-100) coated PEGylated magnetic carbon nanoparticles (denoted as MCM@PEG-CO-DOX NPs) as theranostics nanoplatforms for near-infrared (NIR)-responded CO-DOX combination therapy. MIL-100 as a good nanocarrier of DOX with high loading capacity can also chelate the Mn carbonyl after a smart modification. Meanwhile, magnetic carbon core possessed photothermal effect, which can convert the NIR light to heat by an 808 nm laser irradiation, resulting in the on-demand release of CO and DOX. As a result, combining with PTT, MCM@PEG-CO-DOX NPs killed tumor efficiently. Moreover, our synthesized MCM@PEG-CO-DOX NPs were capable of realizing tumor dual-mode imaging including magnetic resonance imaging (MRI) and photoacoustic imaging (PAI).

  • Polydopamine-Based Tumor-Targeted Multifunctional Reagents for Computer Tomography/Fluorescence Dual-Mode Bioimaging-Guided Photothermal Therapy

    摘要: Development of multifunctional diagnosis and treatment reagents is very meaningful in clinical application. Herein, we developed a polydopamine-based (PDA-based) tumor targeted multifunctional reagent by surface-initiated atom transfer radical polymerization (ATRP) strategy. First, the targeted PDA nanoparticles were prepared via combining with folic acid (FA) and dopamine. Then ATRP technology was used to graft the europium(III) complexes onto PDA surface (defined as FEDA). A series of detections revealed that the FEDA nanoparticles had been successfully prepared and exhibited a bright X-ray computer tomography (CT) and photoluminescence (PL) dual-mode imaging efficiency and an excellent photothermal therapy (PTT) effect in vivo/in vitro.

  • Multifunctional Ag@NaGdF4:Yb3+, Er3+ core-shell nanocomposites for dual-mode imaging and photothermal therapy

    摘要: Multifunctional core-shell nanostructure, which can be applied on bio-imaging and photothermal therapy (PTT), has attracted tremendous interest in recent years. Here, Ag@NaGdF4:Yb3+, Er3+ core-shell nanostructure incorporating luminescent, magnetic and photothermal conversion properties was prepared via a simple method. The as-prepared nanocomposites (NCs) exhibit uniform core-shell sphere morphology, green up-conversion luminescence (UCL), good paramagnetic and photothermal conversion behaviors. The luminescence resonance energy transfer (LRET) between Ag nanoparticles (NPs) and NaGdF4:Yb3+, Er3+ NPs were researched. The cytotoxicity test on HeLa cells using methyl thiazolyltetrazolium (MTT) assay shows good biocompatibility of the core-shell NCs. Moreover, in vitro computed tomography (CT) and UCL imaging studies and in vitro PTT of living assay suggest that the core-shell NCs can be used as theranostic nanoagents for dual-mode imaging guided PTT for tumor.
