Aqueous synthesis of glutathione-capped CuInS2/ZnS quantum dots-graphene oxide nanocomposite as fluorescence a??switch OFFa?? for explosive detection
摘要: This work reports a simple and fast aqueous preparation of CuInS2/ZnS-graphene oxide (CIS/ZnS-GO) nanocomposite as a fluorescence "switch OFF" probe for the fluorescence detection of 2, 4, 6-trinitrophenol (TNP.) – a raw material for various explosive devices. The as-synthesized nanocomposites was characterised using Ultraviolet–visible (UV–Vis) spectroscopy, photoluminescence (PL) spectroscopy, high resolution transmission electron microscopy (HR-TEM), Fourier-transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD) and Raman scattering. The PL studies revealed that the adsorption of TNP onto GO via p-p stacking enhanced the charges transfer from CuInS2/ZnS-GO to the analyte. The limit of detection (LOD) for the analyte is 57 lΜ.

Fluorometric determination of glucose based on a redox reaction between glucose and aminopropyltriethoxysilane and in-situ formation of blue-green emitting silicon nanodots
摘要: A method is described for fluorometric detection of glucose. It is based on the finding that silicon nanodots (SNDs) are formed from glucose and aminopropyltriethoxysilane (APTES) under mild experimental conditions. The SNDs thus formed have an average diameter of ~2 nm, exhibit good water dispersibility, blue fluorescence (with excitation/emission maxima at 410/475 nm), broad pH tolerance, and are photostable. The assay was applied to the quantification of glucose with high sensitivity, good specificity, and over a wide detection range (from 10 μM to 0.9 mM). It was applied to the determination of glucose in spiked serum samples and gave satisfactory results and recoveries.

Amino-Functionalized Al-MOF for Fluorescent Detection of Tetracyclines in Milk
摘要: A fluorescent method for detection of tetracyclines (TCs) in milk was developed by using the NH2-MIL-53(Al) nanosensor synthesized via a one-pot hydrothermal method. The nanosensor had a crystalline nanoplates structure with rich groups of -NH2 and -COOH. The -NH2/-COOH of NH2-MIL-53(Al) reacted with the -CO-/-OH of TCs to form a complex. The electron of -NH2/-COOH from the NH2-BDC ligand transferred to the -CO-/-OH of TCs. -NH2 of the NH2-MIL-53(Al) interacted with the -CO-/-OH of TCs by hydrogen bonding. Quenching efficiency of inner filter effect (IFE) was calculated to contribute 57-89%. The synergistic effect of photoinduced electron transfer (PET) and IFE account for fluorescence quenching. TCs were quantitatively detected in milk samples with recoveries of 85.15 ~ 112.13%, the results were in great accordance with high performance liquid chromatography (HPLC) (P > 0.05), confirming the NH2-MIL-53(Al) nanosensor has potential applicability for the detection of TCs in food matrix.

Operando observation of chemical transformations of iridium oxide during photoelectrochemical water oxidation
摘要: Iridium oxide is one of the few catalysts capable of catalyzing the oxygen evolution reaction (OER) in both acidic and basic conditions. Understanding the mechanism of IrOx under realistic photoelectrochemical conditions is important for the development of integrated water splitting systems. Herein, we have developed a highly efficient OER photoanode in pH 1 aqueous solutions based on a sputtered IrOx film and a p+n-Si light absorber, interfaced with sputtered Au layer. Operando high energy resolution fluorescence detection X-ray absorption spectroscopy (HERFD XAS) was employed to monitor the oxidation state changes of IrOx during both electrochemical and photoelectrochemical (PEC) water oxidation reactions in pH 1 aqueous solutions. We observed a gradual increase of the average oxidation state of Ir with increasing anodic potential in the pre-catalytic region, followed by a reduction of Ir under O2 evolution conditions. Consistent results were obtained on dark anodes and illuminated photoanodes. However, when the thickness of IrO2 was increased to 2 and 3 nm, the spectral changes became much less pronounced and the reduction of Ir oxidation state after the OER onset was not observed. This is due to the lower surface to bulk ratio, where lattice oxygen sites in the bulk are not accessible for the formation of hydroxide. More generally, the operando method developed here can be extended to other materials, thereby providing a powerful tool for mechanism discovery and an enabling capability for catalyst design.

A dsDNA-lighted fluorophore for monitoring protein-ligand interaction through binding-mediated DNA protection
摘要: Because of their important roles in cellular functions, life activities, drug screening, and disease treatment, the development of efficient methods for monitoring protein-ligand interaction is essential. In this study, inspired by our previous studies on DNA conformation-selective fluorescent indicators, we developed a new sensing platform for monitoring protein-ligand interaction and detecting protein activity based on binding-mediated DNA protection and the dsDNA-lighted fluorophore, ethyl-4-[3,6-bis(1-methyl-4-vinylpyridium iodine)-9H-carbazol-9-yl)] butanoate (EBCB). The ligand was purposefully linked to the 3?-terminal of a hairpin DNA probe to selectively bind with the target protein and protect the DNA from cleavage by exonuclease III. By virtue of EBCB’s outstanding capacity to discriminate DNA conformation, the protein-ligand interaction could be effectively monitored through a fluorescence change in EBCB. A high fluorescence signal was detected when the hairpin DNA was protected in the presence of the target protein, whereas a much lower signal was observed in the presence of nontarget proteins. Our results demonstrated that the proposed strategy had high potential, such as high selectivity and relatively high sensitivity, for monitoring protein-ligand interaction and detecting protein activity. We believe these results will pave the way for applying dsDNA-lighted fluorophore EBCB as an effective signal transducer for DNA conformation transformation-mediated biochemical sensing.

Yellow-emissive carbon dots with a large Stokes shift are viable fluorescent probes for detection and cellular imaging of?silver ions and glutathione
摘要: Yellow-emissive carbon dots (Y-CDs) were prepared by a solvothermal method using anhydrous citric acid and 2,3-phenazinediamine as the starting materials. The Y-CDs display a 24% fluorescence quantum yield, a 188-nm Stokes’ shift and excellent stability. They are shown here to be excellent fluorescent probes for the determination of Ag(I) ion and glutathione (GSH). If exposed to Ag(I) ions, they are bound by the carboxy groups of the Y-CDs, and this causes quenching of fluorescence (with excitation/emission maxima at 380/568 nm) via a static quenching mechanism. This effect was used to design a fluorometric assay for Ag(I). The quenched fluorescence of the Y-CDs can be restored by adding GSH due to the high affinity of GSH for Ag(I). The calibration plot for Ag(I) is linear in the 1–4 μM Ag(I) concentration range, and the limit of detection is 31 nM. The respective values for GSH are 5–32 μM, and 76 nM, respectively. The method was applied to the detection of Ag(I) in spiked environmental water samples and gave recoveries ranging from 93 to 107%. It was also applied to the determination of GSH in tomatoes and purple grapes and gave satisfactory recoveries. The Y-CDs display low cytotoxicity and were successfully used to image Ag(I) and GSH in H1299 cells.

Fluorescence detection test by black printed circuit board based microfluidic channel for polymerase chain reaction
摘要: This paper proposes the optimal structure of a PCB-based micro PCR chip constructed on a PCB substrate using commercial adhesive tapes and plastic covers. The solder mask of the PCB substrate was coated black, and the area where the reaction chamber is attached was legend printed with white silk to minimize the noise during fluorescence detection. The performance of the PCR and fluorescence detection was compared using 6 types of reaction chambers, each made with different double-sided tapes. Three of the chambers were unsuccessful in completing the PCR. The performance of the other three chambers that successfully amplified DNA was compared using Taqman probe for Chlamydia Trachomatis DNA. The amplified product was illuminated diagonally with a blue LED to excite the product just before imaging, and the LED was turned off when the image was captured to prevent quenching of the probe. The images were taken 10 seconds prior to the last extension step for each cycle using a DSLR camera. The experiments were run as a quartet for each three chambers made with different double-sided tape. The results showed that there were significant difference between the three tapes.

Online Third-Order Liquid Chromatographic Data with Native and Photoinduced Fluorescence Detection for the Quantitation of Organic Pollutants in Environmental Water
摘要: Third-order liquid chromatographic data were generated online for the simultaneous quantitation of six organic environmental pollutants. The employed strategy consists in reducing the linear flow rate at the column outlet. A postcolumn UV reactor and a fluorimetric detector allowed to properly record both photoinduced and native excitation?emission fluorescence matrices (EEPIFMs and EEFMs, respectively). The obtained third-order liquid chromatography data were chemometrically processed with the multivariate curve resolution?alternating least-squares model. The sensitivity of the overall analytical method was enhanced by a very simple solid-phase extraction with C18 membranes, to be able to successfully apply it to natural water samples tested as real matrices. Favorable detection limits for the investigated pollutants, ranging from 0.02 to 0.27 ng mL?1, were attained, with relative prediction errors between 2 and 7%. Since the studied samples contain uncalibrated interferents, the applied strategy achieves the second-order advantage. Implications regarding the potential achievement of the third-order advantage are discussed.

Fe3+-selective and sensitive “on-off” fluorescence probe based on the graphitic carbon nitride nanosheets
摘要: An effective and facile “on-off” fluorescence sensing approach for the determination of Fe3+ ion using a large area and relatively uniform size graphitic carbon nitride nanosheets (GCNS) was developed. The prepared GCNS have blue and stable emission, as well as excellent water dispersion, and were applied as an effective fluorescent probe that based on the quenched fluorescence for selective and sensitive detection of Fe3+ ion. Herein, we explain the ambiguous fluorescence quenching mechanism between the GCNS and Fe3+, which mainly springs from the redox potential and empty d orbital of Fe3+. The redox potential and unfilled d orbit of Fe3+ endow it excellent binding force with GCNS, which generates most obvious fluorescence quenching effect with respect to other metal ions. The limit of detection (LOD) for Fe3+ was found to be about 2.06 μM. Therefore, the prepared GCNS has the potential to be used as a fluorescent probe for detection.

A practical method for preparing fluorescent-labeled glycans with a 9-fluorenylmethyl derivative to simplify a fluorimetric HPLC-based analysis
摘要: Analysis of glycans in glycoproteins is often performed by liquid chromatography (LC) separation coupled with fluorescence detection and/or mass spectrometric detection. Enzymatically or chemically released glycans from glycoproteins are usually labeled by reductive amination with a fluorophore reagent. Although labeling techniques based on reductive amination have been well-established as sample preparation methods for fluorometric HPLC-based glycan analysis, they often include time-consuming and tedious purification steps. Here, we reported an alternative fluorescent labeling method based on the synthesis of hydrazone and its reduction using 9-fluorenylmethyl carbazate (Fmoc-hydrazine) as a fluorophore reagent. Using isomaltopentaose and N-glycans from human IgG, we optimized the Fmoc-labeling conditions and purification procedure of Fmoc-labeled N-glycans and applied the optimized method for the analysis of N-glycans released from four glycoproteins (bovine RNase B, human fibrinogen, (cid:2)1-acid glycoprotein, and bovine fetuin). The complete workflow for preparation of fluorescent-labeled N-glycans takes a total of 3.5 h and is simple to implement. The method presented here lowers the overall cost of a fluorescently labeled N-glycan and will be practically useful for the screening of disease-related glycans or routine analysis at an early stage of development of biopharmaceuticals.
