  • Track-etched membrane as fluorescence-based pH biosensor

    摘要: Robust and cost-effective stimuli-responsive polymers show prominent advantages to be integrated into detection devices. In addition, modified track-etched membranes with chemical-sensing polymers possess additional robustness features such as including the sensing material into sub-micron pores. In this study, we report the preparation of track-etched PET membranes with fluorescent response in direct relation to changes in the pH of the environment. Immobilised Fluorescein and Green Fluorescent Protein have been used as pH-sensor elements. The former was not sensitive to pH, while the latter had a similar pH sensitivity to that of the free protein. Modifications of track-etched membranes were carried out by grafting polymerisation initiated by the remnant radicals, a straightforward technique for selective modification of the inner wall of pores. The biosensor prepared with the fluorescent protein was able to sense the pH of a buffer solution in the range 4 to 8. Furthermore, this membrane evidenced capacity to sense the pH of the cell growth by in situ fluorescence intensity detection during E.coli cell culture in microwells.

  • Plasmonic oxidation of glycerol using AuPd/TiO2 catalysts

    摘要: AuPd nanoparticles supported on P25 TiO2 (AuPd/TiO2) were prepared by a facile sol-immobilisation method and investigated for surface plasmon-assisted glycerol oxidation under base-free conditions. The AuPd/TiO2 samples were characterized by UV-vis spectroscopy and transmission electron microscopy. The sol-immobilisation method readily permitted the Au:Pd molar ratio to be changed over a wide range whilst keeping the mean particle size of the AuPd nanoparticles at 3nm. Visible light irradiation during the reaction has a beneficial effect on the conversion of glycerol with the most marked effect being observed with gold-rich catalysts and the increase of conversion on light irradiation increases linearly with the gold content of the nanoparticles. The reaction selectivity is also affected by the plasmon-assisted oxidation and glycolic acid, not observed during the dark reactions, was observed for all illuminated reactions due to the enhanced activity of these catalysts.

  • Rapid immunofluorescence assay for staphylococcal enterotoxin A using magnetic nanoparticles

    摘要: A competitive immunoassay for staphylococcal enterotoxin A (SEA) detection in milk was developed, using immobilised antibody onto magnetic nanoparticles (MNPs). MNPs were prepared and then modified to introduce amino groups on them. The morphology and size of the obtained both unmodified and modified MNPs were characterized using TEM analyses. Monoclonal anti-SEA antibody was immobilised onto the modified MNPs (MNP-Ab). Staphylococcal enterotoxin A was conjugated with fluorescent dye ATTO620NHS. The characteristics of fluorescence conjugate were examined. The amount of MNP-Ab and concentration of the fluorescent conjugate used for competitive immunoassay were optimized: 0.25 mg and 53 lg mL(cid:1)1, respectively. The detection limit of developed immunoassay was determined – 0.23 ng mL(cid:1)1 SEA in spiked milk samples. The immunoassay takes only 30 min, the magnetic separation is fast (<10 s) and the volume of the sample for analysis is very small (200 lL).

  • A comprehensive review on thin film-based nano-biosensor for uric acid determination: arthritis diagnosis

    摘要: This article aims to provide a survey on the development of thin film-based nano-biosensor over the decade, which includes sensing principles, electrode types and various thin film matrices. The theoretical and experimental results suggest that the thin film-based sensor are suitable for highly sensitive detection of low concentration bio molecules and can be used for early diagnosis and screening of arthritis, kidney diseases and other cardiovascular diseases. This review concludes that enzyme immobilisation and surface functionalisation methods were used to increase sensitivity of the biosensor and new combination of polymer nano-composites with metal oxide thin films results in increased efficiency due to its high biocompatibility.
