Fluorescent membrane markers elucidate the association of Borrelia burgdorferi with tick cell lines
摘要: This study aimed to describe the association of Borrelia burgdorferi s.s. with ixodid tick cell lines by flow cytometry and fluorescence and confocal microscopy. Spirochetes were stained with a fluorescent membrane marker (PKH67 or PKH26), inoculated into 8 different tick cell lines and incubated at 30°C for 24 h. PKH efficiently stained B. burgdorferi without affecting bacterial viability or motility. Among the tick cell lines tested, the Rhipicephalus appendiculatus cell line RA243 achieved the highest percentage of association/internalization, with both high (90%) and low (10%) concentrations of BSK-H medium in tick cell culture medium. Treatment with cytochalasin D dramatically reduced the average percentage of cells with internalized spirochetes, which passed through a dramatic morphological change during their internalization by the host cell as observed in time-lapse photography. Almost all of the fluorescent bacteria were seen to be inside the tick cells. PKH labeling of borreliae proved to be a reliable and valuable tool to analyze the association of spirochetes with host cells by flow cytometry, confocal and fluorescence microscopy.

摘要: A landing system is an essential part of an Autonomous Unmanned Aerial Vehicle (UAV) to perform autonomous aviation missions. Such a system requires a landing mechanism and a procedure that will allow a safe return to a selected landing position when the mission tasks are finished. In this paper the authors present a concept and first results of a developed system that is connected to a drone which allows automatic landing of a vehicle based on image processing of images captured in real-time by on-board cameras. The system consists of three different cameras: a visible light camera, an infrared camera and a thermal imaging camera. Preliminary results and comparison of developed image processing algorithms for different cameras and two different landing markers are also presented here.

3D GaN Fins as a Versatile Platform for a-Plane-Based Devices
摘要: GaN fins on GaN-on-sapphire templates are fabricated by continuous mode selective area metalorganic vapor phase epitaxy. The fins exhibit high aspect ratios and smooth nonpolar a-plane sidewalls with an ultra-low threading dislocation density of a few 105 cm?2 making them ideally suited for optoelectronic to electronic applications. A detailed analysis of the inner structure of GaN fins is provided by the help of marker layer experiments and correlation of results from fins fabricated under different growth conditions, leading to the development of a growth model to explain the final geometry and optical as well as electrical properties of these high aspect ratio fins. Distinctly different material properties for the central and outer parts of the fins are detected. Whereas the outer sidewalls represent high quality GaN surfaces with very low defect densities, a strong quenching of near band edge emission (NBE) in the central part of the fins is accompanied by heavy compensation of free electrons. A possible explanation is the incorporation of excessive point defects, like intrinsic defects or carbon impurity. The sidewall regions, however, prove to be highly suitable for device applications due to their strong NBE emission, low dislocation density, and high free carrier concentration.

Synthesis of new zinc (II) phthalocyanine conjugates with block copolymers for cancer therapy
摘要: Synthetic routes towards new conjugates of hydrophilic zinc(II) phthalocyanine (ZnPc) with poly(ethylene oxide)-b-poly(propylene oxide)-b-poly(ethylene oxide) (Pluronic P123) and poly(L-lactide) (PLLA), are described. The main semiproduct ZnPc was obtained by heating 4-nitrophthalimide with urea and zinc chloride, followed by the reduction step. Steglich esterification was used to synthesize two ZnPc-conjugated block copolymers, further utilized in fabrication of polymeric micelles (PMs) - functionalized with the zinc(II) phthalocyanine-type moiety. Biological evaluation of the PMs indicated an acceptable biocompatibility level in accord with requirements in the field of nanotheranostics and nanomedicine.

Convenient Construction of Orthogonal Dual Aptamer-Based Plasmonic Immunosandwich Assay for Probing Protein Disease Markers in Complex Samples and Living Animals
摘要: Aptamers, due to their outstanding merits including simple synthesis and easy modification, have been widely used as antibody alternatives to construct novel immunosandwich assays. Dual aptamer-based sandwich assays exhibit multiple advantages over conventional immunosandwich assays and single aptamer-based sandwich assays. However, their construction is hampered by the limited knowledge of binding orthogonality of aptamers reported in literature. Herein we present a new strategy for conveniently constructing an orthogonal dual aptamer-based plasmonic immunosandwich assay (odA-PISA) for probing proteins in complex samples and living animals. An orthogonal aptamer pair was first efficiently selected from aptamers reported in literature by affinity capillary electrophoresis (ACE). Then, a target protein-capturing gold thin layer-coated probe and silver nanoparticle-based Raman labeling nanotags were conveniently prepared with the selected aptamers and used to construct the assay. The double aptamers used ensured the specificity while the plasmonic coupling effect between the target-capturing probe and Raman nanotags generated significantly enhanced Raman signal intensity, providing high sensitivity. As a proof of principle, alkaline phosphatase (ALP) was used as the target. The constructed odA-PISA exhibited high specificity and high sensitivity toward ALP, giving cross-reactivity ≤ 4.2% and limit of detection (LOD) of 3.8 pM (S/N = 4). Quantitative determination of ALP in human serum and probing ALP in tumor-bearing mice were achieved, showing the great application potential of the method. This strategy is widely applicable to other protein disease markers. Therefore, it opened a new access to the construction of sensitive dual aptamer-based sandwich assays for real-world applications particularly disease diagnosis.

The influence of He-Ne laser on agro-morphological criteria, ISSR marker and SDS-PAGE of Moringa oleifera
摘要: Background: Moringa oleifera L. (moringa) is a promising plant for nutritionally and medicinally uses. The influence of laser radiation on moringa seeds (dry and wetted) was investigated, using helium–neon (He–Ne) laser source at ? = 632.8 nm wavelength and 5 cm beam diameter for 25, 45, and 90 min. Agro-morphological criteria, SDS-PAGE protein banding patterns, and ISSR markers were investigated to elucidate the influence of He–Ne laser on moringa. Results: Most agro-morphological criteria increased especially with wetted seed group pre-laser treatments (25 min). SDS-PAGE and ISSR profiles showed changes which include the absence of some bands and the appearance of few novel bands. ISSR markers showed a change in band number in all investigated samples with a total number of 172 bands. The polymorphic bands were 91 bands with 19 unique bands. The average mean percentage of polymorphism was recorded 52.91% at samples treated with a laser at different times. Wetted seed treatment for 25 min recorded 40 bands as new bands. Genetic template stability (GTS) values were recorded in all treatments. The lowest GTS % was recorded 76.74 at 25 min (wetted seeds) while highest GTS % was recorded 83.72 at 25 min (dry seeds). According to the data of GTS, produced by laser treatment for 25 and 45 min for wetted and dry seeds, respectively, it is more effective on genome stability than other treatments. Conclusion: He–Ne laser treatments of wetted seeds achieve biostimulation in many aspects of agro-morphological criteria. ISSR analysis is a highly sensitive method for the detection of DNA alteration induced by laser treatments. The exposure of dry and wetted seeds to He–Ne laser at different times affected the genomic template stability percentage (GTS %). Also, the laser treatments have a great effect on gene expression by switching on or off some genes that may lead to the presence or absence of certain proteins, respectively.

Fluorometric visualization of mucin 1 glycans on cell surfaces based on rolling-mediated cascade amplification and CdTe quantum dots
摘要: A rolling-mediated cascade (RMC) amplification strategy is described for improved visualization of profiling glycans of mucin 1 (MUC 1) on cell surfaces. CdTe quantum dots (QDs) are used as fluorescent labels. The RMC based amplification allows even distinct glycoforms of MUC1 to be visualized on the surface of MCF-7 cell via an amplified F?rster resonance energy transfer (FRET) imaging strategy that works at excitation/emission wavelengths of 345/610 nm. This is achieved by utilizing antibody against MUC1 modified with the fluorescent label 7-amino-4-methylcoumarin-3-acetic acid (AMCA) as the energy donor in FRET. The QDs (used to label surface glycans) act as acceptors. N-Azidoacetylgalactosamine-Acetylated (Ac4GalNAz) as a non-natural azido sugar, can be incorporated into the glycans of the cell surface, which can promote further labeling. The method has the advantage of only requiring a small amount of non-natural sugar to be introduced in metabolic glycan labeling since too much of an artificial sugar will interfere with the physiological functions of cells.

AR Enabled IoT for a Smart and Interactive Environment: A Survey and Future Directions
摘要: Accompanying the advent of wireless networking and the Internet of Things (IoT), traditional augmented reality (AR) systems to visualize virtual 3D models of the real world are evolving into smart and interactive AR related to the context of things for physical objects. We propose the integration of AR and IoT in a complementary way, making AR scalable to cover objects everywhere with an acceptable level of performance and interacting with IoT in a more intuitive manner. We identify three key components for realizing such a synergistic integration: (1) distributed and object-centric data management (including for AR services); (2) IoT object-guided tracking; (3) seamless interaction and content interoperability. We survey the current state of these respective areas and herein discuss research on issues about realizing a future smart and interactive living environment.

Hybrid 18F-FDG-PET/MRI Measurement of Standardized Uptake Value Coupled with Yin Yang 1 Signature in Metastatic Breast Cancer. A Preliminary Study
摘要: Purpose: Detection of breast cancer (BC) metastasis at the early stage is important for the assessment of BC progression status. Image analysis represents a valuable tool for the management of oncological patients. Our preliminary study combined imaging parameters from hybrid 18F-FDG-PET/MRI and the expression level of the transcriptional factor Yin Yang 1 (YY1) for the detection of early metastases. Methods: The study enrolled suspected n = 217 BC patients that underwent 18F-FDG-PET/MRI scans. The analysis retrospectively included n = 55 subjects. n = 40 were BC patients and n = 15 imaging-negative female individuals were healthy subjects (HS). Standard radiomics parameters were extracted from PET/MRI image. RNA was obtained from peripheral blood mononuclear cells and YY1 expression level was evaluated by real time reverse transcription polymerase chain reactions (qRT-PCR). An enzyme-linked immuosorbent assay (ELISA) was used to determine the amount of YY1 serum protein. Statistical comparison between subgroups was evaluated by Mann-Whitney U and Spearman’s tests. Results: Radiomics showed a signi?cant positive correlation between Greg-level co-occurrence matrix (GLCM) and standardized uptake value maximum (SUVmax) (r = 0.8 and r = 0.8 respectively) in BC patients. YY1 level was signi?cant overexpressed in estrogen receptor (ER)-positive/progesteron receptor-positive/human epidermal growth factor receptor2-negative (ER+/PR+/HER2-) subtype of BC patients with synchronous metastasis (SM) at primary diagnosis compared to metachronous metastasis (MM) and HS (p < 0.001) and correlating signi?cantly with 18F-FDG-uptake parameter (SUVmax) (r = 0.48). Conclusions: The combination of functional 18F-FDG-PET/MRI parameters and molecular determination of YY1 could represent a novel integrated approach to predict synchronous metastatic disease with more accuracy than 18F-FDG-PET/MRI alone.

Diagnosis of the surface morphology of laser-ablated materials using the weighted-DCT approach in laser speckle interferometry for application to plasma facing materials
摘要: A Gram-staining-positive, motile and short-rod-shaped actinobacterium designated 9W16Y-2T was isolated from surface-sterilized leaves of reed (Phragmites australis) collected from Taklamakan Desert in Xinjiang Uygur Autonomous Region, China. Colonies were pale greenish yellow, circular, smooth, and convex. The 16S rRNA gene sequence of strain 9W16Y-2T exhibited highest sequence similarities with Aeromicrobium camelliae CGMCC 1.12942T (99.0%) and Aeromicrobium erythreum NRRL B-3381T (97.2%). Phylogenetic analyses based on 16S rRNA gene sequences and single-copy phylogenetic marker genes (pMGs) showed that strain 9W16Y-2T belonged to the genus Aeromicrobium and formed a monophyletic clade with Aeromicrobium camelliae CGMCC 1.12942T. Furthermore, average nucleotide identity (ANI) and DNA-DNA hybridization (DDH) clearly separated strain 9W16Y-2T from the other species of the genus Aeromicrobium with values below the thresholds for species delineation. The G+C content of the genomic DNA is 68.9 mol%. The diagnostic diamino acid of the cell-wall peptidoglycan was LL-diaminopimelic acid. The predominant menaquinone was MK-9(H4). The major fatty acids (> 10% of the total fatty acids) were C18:0 10-methyl (TBSA) (28.2%), C16:0 (21.0%), C16:0 2-OH (20.8%) and C18:1 ω9c (12.8%). The polar lipid profile comprised diphosphatidylglycerol, phosphatidylglycerol, phosphatidylcholine, phosphatidylinositol, an unidentified aminophospholipid and an unidentified lipid. Based on the phylogenetic, phenotypic and chemotaxonomic features, strain 9W16Y-2T represents a novel species of the genus Aeromicrobium, for which the name Aeromicrobium endophyticum sp. nov. is proposed. The type strain is 9W16Y-2T (= CGMCC 1.13876T = JCM 33141T).
