GE QDs, especially amino-GE QDs, showed excellent signal enhancement for amiodarone and cell components in ToF-SIMS analysis. This strategy holds great potential for further investigation of drug metabolism pathways and the interactions between the cell and micro-environment.

The main problem limiting the wide biological applications of ToF-SIMS is the relatively poor ionization yield of molecules, especially biomolecules, due to hard ionization. The complex chemical micro-environment in single cells typically causes severe matrix effects, leading to significant signal suppression of biomolecules.
1:Experimental Design and Method Selection:
The study compared the ToF-SIMS signal increase for amiodarone by functionalized GE QDs and previously reported signal enhancement materials. GE QDs were further coated on or fed into amiodarone-treated cells to enhance the secondary ion signal of drug, cell membrane-related components, and intracellular components.
2:Sample Selection and Data Sources:
Amiodarone was used as a drug model. Breast cancer MCF-7 cells treated with amiodarone were used as a model sample.
3:List of Experimental Equipment and Materials:
ToF-SIMS V spectrometer, transmission electron microscope (TEM), vacuum freeze drier, UV–vis spectrophotometer, fluorescence spectrophotometer, inverted microscope.
4:Experimental Procedures and Operational Workflow:
Preparation of glass slides and silicon wafers, preparation of analyte-matrix mixtures samples, cell culture, pre-treatment of cell samples, ToF-SIMS instrumentation.
5:Data Analysis Methods:
Positive ToF-SIMS spectra and chemical mapping of single cells were recorded under delayed extraction mode. All data used for quantification was recorded under static limit.
