The study confirms that docking strands in DNA-PAINT microscopy are depleted by irradiation with visible light in an indirect process mediated by the excitation of fluorescent dyes. Two strategies were evaluated to reduce this depletion: the use of oxygen scavenging buffers and increasing the distance between the fluorescent dye and the docking strand. Both strategies showed effectiveness in reducing the depletion of binding sites, with oxygen scavenging buffers being particularly effective at high irradiances.
