研究目的
To examine spatial changes in rod-mediated function in relationship to local structural changes across the central retina in eyes with a spectrum of age-related macular degeneration (AMD) disease severity.
研究成果
Three distinct rod function phenotypes (RF1, RF2, RF3) were identified, with RITslope providing a sensitive measure of spatial changes in dark adaptation. These findings enhance understanding of AMD progression and may inform future therapeutic interventions and outcome measures.
研究不足
Relatively small sample sizes for AMD groups; control group defined by absence of large drusen may include early AMD cases; some participants did not reach criterion threshold for RIT, limiting data completeness.
1:Experimental Design and Method Selection:
Participants were categorized into five AMD severity groups based on fundus features. Scotopic thresholds and dark adaptation were measured using psychophysical methods, and structural changes were evaluated with optical coherence tomography (OCT).
2:Sample Selection and Data Sources:
42 participants aged 53-88 years were recruited from the National Eye Institute clinic, with specific inclusion and exclusion criteria based on AMD severity and ocular health.
3:List of Experimental Equipment and Materials:
Medmont Dark Adapted Chromatic (DAC) perimeter for scotopic thresholds and dark adaptation, ColorDome ganzfeld for pupil measurement and bleaching, spectral-domain optical coherence tomography (SD-OCT) for structural imaging.
4:Experimental Procedures and Operational Workflow:
After pupil dilation, scotopic thresholds were measured at 14 retinal loci after 20 minutes of dark adaptation. A 30% bleach was applied using a full-field background light, followed by dark adaptation measurements at 8 loci over 30 minutes. Structural assessments were done using SD-OCT scans.
5:Data Analysis Methods:
Scotopic thresholds were analyzed in log sc cd/m2, RIT was derived from dark adaptation curves, and RITslope was calculated from linear fits. Statistical analyses included ANOVA and post-hoc tests to compare groups.
