1：Experimental Design and Method Selection:

The study designed PTX-R837-IR820@TSL complexes using a standard thin film/extrusion method. Methods included dynamic light scattering (DLS), transmission electron microscopy (TEM), high-performance liquid chromatography (HPLC), UV-vis spectrophotometry, photothermal performance testing, singlet oxygen detection with SOSG, in vitro cell viability assays (MTT), confocal microscopy for cellular uptake, in vivo biodistribution imaging, and in vivo antitumor efficacy evaluation in tumor-bearing mice.

2：Sample Selection and Data Sources:

MFC mouse gastric carcinoma cells from ATCC were used for in vitro studies. BALB/C mice (6-8 weeks old) were used for in vivo experiments, with tumors induced by subcutaneous injection of MFC cells. Materials included phospholipids (DPPC, DPPG, DSPE-PEG2000), IR820, R837, PTX, and other reagents from suppliers like A.V.T, Aladdin, and Sigma-Aldrich.

4：Experimental Procedures and Operational Workflow:

Liposomal complexes were prepared by dissolving lipids in chloroform/methanol, adding drugs, forming thin films, hydrating with IR820 solution, extruding through polycarbonate membranes, and purifying via gel chromatography. Characterization involved size measurement, morphology imaging, drug release studies at different temperatures and under NIR irradiation, photothermal and singlet oxygen generation tests, in vitro cell viability assays with MTT, cellular uptake imaging, in vivo biodistribution via NIR fluorescence imaging, and in vivo antitumor efficacy with tumor volume measurement and survival analysis.

5：Data Analysis Methods:

Data were analyzed using statistical methods including one-way ANOVA with posthoc Dunnett's test in SPSS software. Cell viability was calculated from absorbance measurements, and cytokine levels were quantified using ELISA. Results are presented as mean ± standard deviation from triplicate or quadruplicate experiments.
