  • Fluorescence microscope light source stability

    摘要: The process of fluorescence starts with the efficient generation of light that is required for the excitation of fluorophores. As such, light sources are a crucial component of a fluorescence microscope. Choosing the right illumination tool can not only improve the quality of experimental results, but also the microscope’s economic and environmental footprint. While arc lamps have historically proven to be a reliable light source for widefield fluorescence microscopy, solid-state light-emitting diodes (LEDs) have become the light source of choice for new fluorescence microscopy systems. In this paper, we demonstrate that LEDs have superior light stability on all timescales tested and use less electrical power than traditional light sources when used at lower power outputs. They can be readily switched on and off electronically, have a longer lifetime and they do not contain mercury, and thus are better for the environment. We demonstrate that it is important to measure light source power output during warm-up and switching, as a light source’s responsiveness (in terms of power) can be quite variable. Several general protocols for testing light source stability are presented. A detailed life cycle analysis shows that an LED light source can have a fourfold lower environmental impact when compared to a metal halide source.

  • Mercury Free Microscopy: An Opportunity for Core Facility Directors

    摘要: Mercury Free Microscopy (MFM) is a new movement that encourages microscope owners to choose modern mercury free light sources to replace more traditional mercury based arc lamps. Microscope performance is enhanced with new solid state technologies because they offer a more stable light intensity output and have a more uniform light output across the visible spectrum. Solid state sources not only eliminate mercury but also eliminate the cost of consumable bulbs (lifetime (cid:2)200 hours), use less energy, reduce the instrument down time when bulbs fail and reduce the staff time required to replace and align bulbs. With lifetimes on the order of tens of thousands of hours, solid state replacements can pay for themselves over their lifetime with the omission of consumable, staff (no need to replace and align bulbs) and energy costs. Solid state sources are also sustainable and comply with institutional and government body mandates to reduce energy consumption, carbon footprints and hazardous waste. MFM can be used as a mechanism to access institutional ?nancial resources for sustainable technology through a variety of stakeholders to defray the cost to microscope owners for the initial purchase of solid state sources or the replacement cost of mercury based sources. Core facility managers can take a lead in this area as “green” ambassadors for their institution by championing a local MFM program that will save their institution money and energy and eliminate mercury from the waste stream. Managers can leverage MFM to increase the visibility of their facility, their impact within the institution, and as a vital educational resource for scienti?c and administrative consultation.

  • Using Alternative Light Source Technology to Enhance Visual Inspection of the Skin

    摘要: BACKGROUND: The identification of deep tissue pressure injury (DTPI) in the early stages of evolution presents a challenge, as skin compromise is only visually apparent when evidence of damage reaches its outer layers. CASE DESCRIPTION: We describe use of an alternative light source (ALS) to enhance visual skin assessment in 3 cases. Case 1 was a 47-year-old African American man with a hyperpigmented inner buttocks and a mixture of partial- and shallow full-thickness skin loss from incontinence-associated dermatitis and friction. Case 2 was a 62-year-old African American woman with a gluteal cleft DTPI. Case 3 was a 57-year-old African American woman with a stage 3 pressure injury of the right buttock. CONCLUSION: The ALS enabled visualization of skin nuances not visible to the unaided eye. Based on this experience, we conclude that use of an ALS provided additional visual details when compared to traditional visual inspection. We found that as the ALS interplays with skin layers, penetrating and absorbing at differing depths, compromised skin appeared darker and more distinct when compared to adjacent, intact skin. Additional research is needed to determine whether the ALS enables earlier pressure injury detection, timelier and more effective intervention, decreased morbidity, and cost savings.

In this article is presented short overview of color rendering indices history with discussion about possible improvement of CIE color rendering index by gamut mapping.

    摘要: We developed rare-earth doped phosphors for mercury-free narrowband ultraviolet light sources. The narrowband emission is a typical feature of the intra-orbital electron transition in the rare-earth Gd3+ ion. Al1-xGdxN epitaxial films were grown by using an ultra-pure reactive sputtering technique performed under molecular-beam epitaxial quality. A narrowband luminescence line from Gd3+ ions has been observed at 318 nm. We fabricated field-emission devices using Al1-xGdxN thin films and investigated detailed device characteristics. Furthermore, recently, we have successfully developed bright fluorescent lamps with an ultraviolet phosphor containing Gd3+ ions.

    摘要: We developed flat panel UV-C light generation technique with the electron beam pumping technology using carbon nanotube (CNT) emitters. The wide bandgap anode material excited with high performance CNT based cold cathode electron beam (C-beam). Flat panel UV-C light generation efficiencies depend on the anode biases and current densities. With the optimized C-beam and anode, we could obtain UV-C with wavelength of 269.5 nm. Flat panel UV-C light source would be applied to many industries because advantages of flat areal light source.

    摘要: Luminous flux (lm) and active electrical power consumption (W) of new SSL products coming to market are measured at test laboratories in order to determine the luminous efficacy (lm/W) and energy class of the products. Due to the fact that spectral responsivities of V(λ)-filtered photometers differ from the defined CIE V(λ) curve, all photometers are typically calibrated using light with correlated colour temperature (CCT) of 2856 K. This causes unwanted spectral errors in the measurements of SSL products at test laboratories and also in the field due to the large differences in the spectra of calibration sources and the light being measured, and in many cases cannot be corrected for. The EMPIR (European Metrology Programme for Innovation and Research) project 15SIB07 PhotoLED addresses the obsolescence of traditional incandescent standard lamp technology used in photometry and the need to support the introduction and uptake of solid-state lighting (SSL) products. It develops and validates the basis for a new photometric system based on white LEDs, by developing new LED-based calibration sources and measurement techniques supporting the specific properties of the new light sources and detector technology including illuminance measurement of the new LED-sources without optical V(λ)-filters in the calibration of photometers and measurement facilities used for determining the energy efficiency of new SSL products coming to market.

    摘要: The last decade is the time of a very dynamic development of lighting technology. This development covers many areas that complement each other. The development of light sources, luminaires, measurement techniques and design methods make it necessary to develop new, effective tools to support the design and analysis of lighting equipment. In lighting technology, the most common analysis is made of such parameters as the illuminance, luminance distribution and discomfort glare, inseparable from the specificity of modern light sources. A characteristic feature of almost all of them is high luminance, on the level of 107 – 108 cd/m2. High luminance facilitates the design of very precise optical systems, but also it is strongly associated with the possibility of discomfort associated with glare. Current methods of glare analysis in simulation software are related to conversion to the average luminance value of the luminaire, saved as simplified light parameters in electronic files (IES, LDT or similar). Currently the calculations take place as a result of calculation ratio the value of luminous intensity in a given direction and the surface area of the dimension of the luminaires in the same direction. This can lead to a significant understatement of the luminance of the luminaire in any situation where luminous intensity comes from the heterogeneous luminance distribution on the surface of the luminaire. This situation most often occurs in luminaires with high-luminance light sources with a reflector system and in the case of multi-source luminaires with lenses - eg multi-LED luminaires. The paper presents advanced methods of luminance modelling of light sources developed as a result of digital measurements of luminance distributions. A comparative analysis of the accuracy of the simulation calculations of the analytical lighting fixture model will be presented. The results will be compared to the classic ray tracing methods used in currently used models.

  • Light source estimation using feature points from specular highlights and cast shadows

    摘要: A method for light sources estimation is proposed in this paper. The method utilizes feature points in cast shadows to estimate near light source positions from estimated source directions using specular highlights. There are several methods that can be used to estimate light sources from scene images, using either cast shadows or specular highlights. However, most of them are limited to directional light sources. The proposed method can estimate the positions and intensities of multiple near point light sources. Specular highlights on an object of known geometry are first used for light source direction estimation. Then, a discontinuity point in the object shape and the corresponding cast shadow on a ground plane are used for light source position estimation. Feature points obtained from an image of the cast shadow, however, can be inaccurate due to various factors. Information on diffused light reflected from Lambertian ground-plane surface is subsequently used to improve estimation accuracy. Experimental results were used to evaluate the performance of the proposed method.
