  • Protective effect of MAAs extracted from Porphyra tenera against UV irradiation-induced photoaging in mouse skin

    摘要: The objective of this study was to investigate the effect of Mycosporine-like amino acids (MAAs) extracted from Porphyra tenera skin against UV irradiation-induced photoaging using an ICR mouse model of skin photoaging and to explore the curative effects of the compounds in MAAs. The skin damage and collagenous tissue impairments induced by ultraviolet radiation were observed by histopathological analysis, and the effects of MAAs on protecting against skin damage and maintaining an intact structure of collagenous tissue were studied. The expression of NF-κB and the MAPK signaling pathway and nuclear transcription factors MMP-1, MMP-3 and TNF-α was analyzed used quantitative real-time reverse transcription-polymerase chain reaction (qRT-PCR) and ELISA assays. The results showed that the MAAs extracted from Porphyra tenera contained Porphyra-334 and shinorine, which could prevent skin photoaging induced by ultraviolet irradiation and reduce the damage to collagen and elastin. Meanwhile, MAAs significantly inhibited the decrease in hydroxyproline and collagen content and protected against pathological damage to collagen fibers in photoaging skin. MAAs resulted in a reduction in the expression of interstitial collagenase (MMP-1), matrix metalloproteinases (MMP-3) and tumor necrosis factor-α (TNF-α) and a reduction in the content of tissue matrix metalloproteinase (MMPs). Furthermore, MAAs may promote procollagen synthesis by downregulating the expression of TNF-α and downregulating the levels of MMPs, which demonstrates that MAAs are involved in matrix collagen synthesis by activating the NF-kB pathway in photoaging skin. Based on these results, we concluded that MAAs protect skin from UV irradiation-induced photodamage, and therefore, MAAs may be a potentially effective agent for the prevention of photoaging.

  • The influence of MC1R dermal morphological features of photoexposed skin in women revealed by reflectance confocal microscopy and optical coherence tomography

    摘要: Background: The melanocortin 1 receptor (MC1R) gene is one of the major determinants of skin pigmentation. It is a highly polymorphic gene and some of its polymorphisms, have been related to specific skin phenotypes, increased risk of skin cancers and skin photoaging. Currently, its contribution to changes in dermal features in photo-exposed skin is unknown. Objective: The main objective of this study is to evaluate the potential correlation between MC1R status and specific healthy photoexposed skin characteristics. Materials and methods: Skin facial features were estimated by evaluation with standard digital photography with automated features, reflectance confocal microscopy (RCM) and optical coherence tomography (OCT) in 100 healthy women. Skin of the forearms was used as a control. Results: The study found an association between RHC MC1R polymorphisms and dermal features in photoexposed areas being represented by increased vessel density and pixel density in OCT (p=0.025 and p=0.001, respectively) and increased coarse collagen in RCM (p=0.034), as compared to non-RHC subjects. To our knowledge this is previously unreported. Additionally, previously reported correlations between light hair and skin color and pigmented spots with MC1R RHC polymorphisms have been confirmed. Conclusions: Our results suggest the role of RHC MC1R variants in dermal variations of facial skin, as compared to non-RHC variants. To our knowledge this is previously unreported.

  • Split-face comparison of the picosecond 1064-nm Nd:YAG laser using a microlens array and the quasi-long-pulsed 1064-nm Nd:YAG laser for treatment of photoaging facial wrinkles and pores in Asians

    摘要: Skin photoaging manifests deeper wrinkles and larger pore size. Various lasers have been tried for rejuvenation of photoaging skin, and the quasi-long-pulsed 1064-nm neodymium:yttrium-aluminum-garnet (Nd:YAG) laser is one promising treatment option. New types of laser device are emerging, including devices operating with picosecond pulse duration combined with a microlens array, which are regarded as a new breakthrough for skin rejuvenation. We aimed to evaluate the clinical effectiveness and safety of the picosecond 1064-nm Nd:YAG laser using a microlens array compared with the quasi-long-pulsed 1064-nm Nd:YAG laser in a split-face design. We designed a split-faced, prospective study and enrolled 25 subjects with photoaging facial wrinkles and enlarged pores. Each facial area was randomly assigned to undergo treatment with either the picosecond 1064-nm Nd:YAG laser (Pico-arm) or the quasi-long-pulsed 1064-nm Nd:YAG laser (Quasi-arm). We performed five laser sessions at 2-week intervals, and final results were assessed after 20 weeks after the initial laser session. We used a five-point global assessment scale, wrinkle and pore index derived from 3D camera analysis. We enrolled a total of 25 subjects (24 females and 1 male) with Fitzpatrick skin types III to IV and a mean age of 63.8 ± 5.7 years. After treatment, 54.2% of subjects in the Pico-arm reported at least moderate improvement in visible pores compared with 41.7% of the Quasi-arm (P = 0.001). A total of 12.5% of subjects in the Pico-arm showed moderate improvement in wrinkles versus 4.2% of the Quasi-arm (P = 0.125). There was a 41.3% reduction in the pore index in the Pico-arm compared to a 3.9% increase in the Quasi-arm (P = 0.048). There was a 16.4% reduction in the wrinkle index in the Pico-arm compared with a 0.5% reduction in the Quasi-arm (P = 0.01). Pain assessment score was higher in the Pico-arm than the Quasi-arm (3.65 ± 1.70 vs 1.28 ± 1.28, P = 0.001). No serious adverse events occurred during the study. Our findings suggest that the picosecond 1064-nm Nd:YAG laser with a microlens array is as effective as the quasi-long-pulsed 1064-nm Nd:YAG laser for treatment of photoaging wrinkles and pores.

  • Clinical factors affecting the efficacy of melasma treatment using the Q-switched 1064 Nd: YAG laser mode in a group of Venezuelan female patients

    摘要: The influence of melasma risk factors on the effectiveness of laser toning treatment in Venezuelan females was investigated. Clinical evaluation was performed (n = 64) including ultrasound and thyroid hormone profile. All patients underwent face exposure with laser toning, eight sessions weekly. VISIA percentiles corresponding to dark brown spots determined before and after treatment were compared by ANOVA. Differences between percentile values before and after treatment for each patient were calculated and expressed as percentages. An improvement of ≥30% was considered as significant. Spearman rank correlations between improvement and clinical parameters were determined. A significant increase (p > .005) of the median of the percentiles in the overall group was observed. However, only 62.5% of the patients exhibited an improvement of ≥30% at the right malar, 73.4% at the frontal and 71.8% at the left malar areas. Hormonal contraception during treatment (right malar: p < .0001 left malar: p = .0035), thyroid disorders (right malar: p < .0001; frontal: p = .011; left malar: p < .0001) and photoaging (right malar: p = .0235; frontal: p = .0237; left malar: p = .0137) were inversely associated to melasma improvement after treatment. Prolonged use of sunscreen improved significantly (right malar: p < .001; frontal: p = .016 and left malar: p = .025) treatment effectiveness.

  • Laser Therapy for Actinic Purpura

    摘要: Laser therapy has been previously reported as successful in the treatment of postprocedural ecchymoses. However, laser therapy has not been formally evaluated in the context of actinic purpura. Actinic purpura, also known as solar purpura, is a relatively common manifestation of photoaging that falls under the broader classification of dermatoporosis, or skin fragility secondary to photoaging. It is estimated that 12% of adults older than 50 years experience actinic purpura, with the proportion increasing to 30% for adults older than 75 years. Although a benign condition, actinic purpura can be an unsightly nuisance for patients, which may even affect their quality of life. With a normal resorption time of approximately 10 to 14 days, this condition may have a continued significant impact on patients given that it can occur with minor or unnoticed trauma. Previous reported attempts at treating and preventing actinic purpura have included topical retinoids, topical human epidermal growth factor, and combination topical formulations.

  • A split-face randomized controlled trial of treatment with broadband light for enlarged facial pores

    摘要: Background: Broadband light (BBL) is one of the intense pulsed light. BBL treatment for nonablative, noninvasive skin tightening and rejuvenation reportedly elicits satisfactory clinical outcomes. Up until now, self-controlled case series with objective evaluation of efficacy of facial pores in response to BBL treatment have not been reported. We aimed to assess its safety and efficacy on enlarged facial pores by objective measuring technique. Objective: To evaluate the efficacy and safety of BBL on the appearance of facial pores. Methods: 25 patients received five treatments with BBL at 2-week intervals. One split-face was treated by BBL, another split-face was untreated as a control. Objective measurements of the quantitative number of pores were calculated with a VISIA-CR imaging system. Skin elasticity, transepidermal water loss, skin sebum excretion, and skin surface hydration values were also evaluated by using an Elastimeter, Vapometer, Sebumscale, and Moisturemeter. Results: Pretreatment, the total average number of facial pores was 835.88 and 835.68 for BBL-treated facial sides and untreated facial sides, respectively (p ? .993). These values were 801.44 and 811.8, respectively, 2 months after the final treatment (p ? .606). There were no long-term side effects. Conclusion: BBL seems to be a safe and mildly effective approach to reduce detectable pores.

  • Case histories of intense pulsed light phototherapy in dermatology - the HPPL? and IFL? technologies

    摘要: The intense pulsed light (IPL) and laser technologies are widely used for skin rejuvenation and for treating several dermatological disorders such as skin dyschromia and acne, and for non-ablative dermal remodeling of rhytides and hypertrophic scars. Technological evolution is rapid. The High Power Pulsed Light? [HPPL?] and Incoherent Fast Light? [IFL?, Novavision Group S.p.A., 20826 Misinto (MB), Italy] are recent innovations in the field of IPL technologies; IFL? is a further evolution of the already advanced HPPL? system. The paper presents a selection of case histories of dermatological lesions treated with the HPPL? and IFL? technologies. All study materials were appropriately peer-reviewed for ethical problems.
