1:Experimental Design and Method Selection:
MG-63 cells were divided into four groups to receive different treatments including doxycycline injection, diode laser radiation, a combination of both, and a control group with daily medium replacement.
2:Sample Selection and Data Sources:
The human osteosarcoma cell line (MG-63) was used, obtained from the cell bank of Royan Institute.
3:List of Experimental Equipment and Materials:
Diode laser (810 nm, 100 mw), doxycycline (2 mg/mL), culture medium containing DMEM, FBS, L-glutamine, and penicillin-streptomycin.
4:Experimental Procedures and Operational Workflow:
Treatments were administered once every 24 h for 4 consecutive days, followed by RNA extraction and cDNA synthesis for measuring OPG and RANKL levels.
5:Data Analysis Methods:
The levels of OPG and RANKL gene expression were assessed by one-way analysis of variance (ANOVA) and post hoc Fisher’s least significant difference (LSD) tests.
