研究目的
Improve mapping and registration of longitudinal view on histopathology vessels in a three-dimensional alignment procedure for postmortem quantitative coronary plaque analyses.
研究成果
The 3D fusion approach enabled slice-based comparison, showing that PMCTA overestimates calcified plaques and OCT underestimates them compared to histopathology. This provides a basis for improving automatic assessment of plaque constitution in future studies.
研究不足
Only two arteries from one heart were included; only 16 OCT images from LAD were of sufficient quality; postmortem settings lack dynamic movement and have deviant pressures, affecting results; annotations were not corrected for errors.
1:Experimental Design and Method Selection:
A 3D alignment procedure was developed to fuse histopathology, PMCTA, and OCT images for quantitative coronary plaque analysis. The procedure involved cropping and aligning histopathology slices, annotating landmarks, and constructing 3D volumes for comparison.
2:Sample Selection and Data Sources:
Data from two coronary arteries (LAD and RCA) from one postmortem heart were used. The heart was placed in a chest phantom (Kyoto Kagaku Lungman).
3:List of Experimental Equipment and Materials:
CT scanner (GE Healthcare CT750 HD), OCT system (Ilumien PCI optimization system from LightLab Imaging, St. Jude Medical), histopathology staining (hematoxylin and eosin), annotation software (QAngio CT Research Edition v
4:1, QCU-CMS-Research v4, custom histopathology annotation program, NPD view2 v1), and various catheters and guidewires. Experimental Procedures and Operational Workflow:
PMCTA and OCT acquisitions were performed, followed by histopathology analysis. Images were processed and annotated independently, then aligned using the 3D fusion procedure with manual landmark matching. Calcium quantification was done semi-automatically.
5:Data Analysis Methods:
Quantitative analysis of calcified plaque areas was performed, comparing modalities using mean areas along the vessel.
